摘 要:目的:探究SHP-2如何调控STAT3-PI3K-AKT信号通路对CNE-1细胞中的活化状态及其对肿瘤生物学行为的影响,并进一步探讨其相关分子机制。方法:采用Western blot检测CNE-1细胞中SHP-2、STAT3、PI3K的表达水平。设CNE-1组为空白组;将空白慢病毒转染至CNE-1细胞后作为NC-SHP-2组;将si-SHP-2病毒转染到CNE-1细胞后作为si-SHP-2组,检测并比较各组SHP-2、STAT3、p-STAT3、PI3K、p-PI3K、AKT、p-AKT的表达水平;平板克隆检测细胞增殖活性;CCK-8检测各组细胞活性;细胞划痕检测各组细胞迁移;Transwell检测各组细胞侵袭;流式细胞术检测各组细胞凋亡;Western blot检测凋亡相关蛋白(Bax、Caspase-3和Bcl-2)的表达水平。结果:SHP-2、STAT3、PI3K在CNE-1细胞中均为高表达。与NC-SHP-2组比较,si-SHP-2组细胞中的p-STAT3,p-PI3K和p-AKT表达显著下调(P<0.05),细胞相对数量、增殖速率、迁移和侵袭速率均显著降低(P<0.05),而细胞凋亡率显著增加(P<0.05)。与NC-SHP-2组比较,si-SHP-2组细胞Bax和Caspase-3的表达水平升高;Bcl-2表达水平降低(P<0.05)。结论:SHP-2在CNE-1细胞中高表达,SHP-2与STAT3-PI3K-AKT通路的激活密切相关,沉默CNE-1细胞中SHP-2的表达能有效降低鼻咽癌的发展。Objective:To explore how SHP-2 affects the activation state of STAT3-PI3K-AKT signaling pathway in nasopharyngeal carcinoma(CNE-1)cells and its influence on tumor biological behavior,and further explore the related molecular mechanisms.Methods:The expression levels of SHP-2,STAT3 and PI3K in CNE-1 cells were detected by Western blot.Set CNE-1 group as blank group,transfecting blank lentivirus into CNE-1 cells as NC-SHP-2 group.The si-SHP-2 virus was added to the CNE-1 cells for the si-SHP-2 group.The expression levels of SHP-2,STAT3,p-STAT3,PI3K,p-PI3K,AKT and p-AKT were detected and compared between the groups.Cell proliferation was detected by plate cloning.CCK-8 was used to detect cell activity.Cell migration was detected by cell scratches.Transwell was used to detect cell invasion.Flow cytometry was used to detect apoptosis.The expression of apoptosis-related proteins(Bax,Caspase-3 and Bcl-2)was detected by Western blot.Results:SHP-2,STAT3 and PI3K were highly expressed in CNE-1 cells(P<0.05).Compared with the NC-SHP-2 group,the expression of P-STAT3,P-PI3K and P-Akt in si-SHP-2 group was down-regulated(P<0.05),the relative cell number,proliferation rate,migration rate and invasion rate were decreased(P<0.05),while the apoptosis rate was increased(P<0.05).Compared with the NC-SHP-2 group,si-SHP-2 the expression of Bax and Caspase-3 in si-SHP-2 group increased,and the expression of Bcl-2 decreased(P<0.05).Conclusion:SHP-2 is highly expressed in CNE-1 cells.SHP-2 is closely related to the activation of the STAT3-PI3K-AKT pathway.Silencing the expression of SHP-2 in CNE-1 cells can effectively reduce the development of nasopharyngeal carcinoma.
