摘 要:目的探究丝切蛋白1(Cofilin1,CFL1)是否与头颈鳞状细胞癌(HNSCC)预后不良及免疫检查点阻断疗法耐受相关。方法下载并分析来自不同公共数据库的HNSCC转录组和临床数据,通过生物信息学分析研究CFL1与HNSCC预后、潜在致病机制、免疫浸润、免疫检查点和免疫治疗反应的关系。结果CFL1与HNSCC患者不良预后相关,进一步分析发现CFL1过表达与免疫细胞浸润相关,参与肿瘤免疫逃逸,并且导致HNSCC患者肿瘤免疫功能障碍和排斥的评分升高。结论CFL1表达升高与HNSCC的不良预后、免疫浸润和免疫检查点逃逸相关,并导致HNSCC患者对免疫查点阻断疗法耐受。Objective To investigate whether Cofilin-1(CFL1)is associated with poor prognosis and immune checkpoint blockade therapy resistance in head and neck squamous cell carcinoma(HNSCC).Methods HNSCC transcriptomic and clinical data from different public databases were downloaded and analyzed.Bioinformatics analysis was performed to study the relationship between CFL1 and HNSCC prognosis,potential pathogenic mechanisms,immune infiltration,immune checkpoints,and immune therapy response.Results CFL1 was associated with poor prognosis in HNSCC patients.Further analysis revealed that CFL1 overexpression was correlated with immune cell infiltration,involved in tumor immune evasion,and led to increased scores of tumor immune dysfunction and exclusion in HNSCC patients.Conclusion Elevated CFL1 expression is associated with poor prognosis,immune infiltration,and immune evasion in HNSCC,leading to resistance to immune checkpoint blockade therapy in HNSCC patients.
