摘 要:本试验采用超声辅助乙醇提取大蝎子草地上部分总多酚,通过单因素试验结果设计Box-Behnken模型,优化大蝎子草地上部分总多酚最佳工艺条件,用DPPH自由基清除率、FRAP法评价大蝎子草地上部分总多酚的抗氧化性。结果显示,乙醇浓度、液固比、超声时间、超声温度的最佳工艺参数分别为75%、50 mL/g、35min、40℃时,总多酚提取率为2.81%,与Box-Behnken模型的预测值相符。抗氧化性试验表明,0.15 mg/mL大蝎子草总多酚提取液DPPH清除率可达75.8%,半数抑制浓度IC_(50)值为0.057 mg/mL,具有一定的抗氧化性,为大蝎子草野生资源的研究和利用提供相应的理论支撑,为新型饲料添加剂的开发提供理论基础。The ultrasonic-assisted ethanol was used to extract the total polyphenols from the aerial part of Girardinia diversifolia in this experiment.Box-Behnken model was designed based on the results of single-factor experiments to optimize the optimal process conditions of total polyphenols from the aerial part of Girardinia diversifolia.DPPH free radical scavenging rate and FRAP method were used to evaluate the antioxidant capacity of total polyphenols from the aerial part of Girardinia diversifolia.Results showed when the optimal process parameters as ethanol concentration,liquid-solid ratio,ultrasonic time and ultrasonic temperature were 75%,50 mL/g,35 min and 40℃,respectively,the extraction rate of total polyphenols was 2.81%,which was consistent with the predicted value of Box-Behnken model.The antioxidant test showed that the DPPH radical-scavenging activity of 0.15 mg/mL total polyphenol from the aerial part of Girardinia diversifolia could reach 75.8%,and the IC_(50) value of the half inhibitory concentration was 0.057 mg/mL,which had certain antioxidant capacity.It provides corresponding theoretical data support for further research and utilization of wild resources of Girardinia diversifolia,and a theoretical basis for the development of new feed additives.
