摘  要:目的探讨Bcl-2相关抗凋亡基因3(BAG3)对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞增殖和迁移能力的影响。方法通过查询并整理TCGA数据库,对不同肿瘤中的BAG 3表达进行泛癌分析。通过蛋白免疫印迹实验检测NSCLC细胞系(H1299、A549、H460、Anip973、PC9、H358和95-D细胞)以及人正常肺上皮细胞系Beas-2B中BAG3蛋白的相对表达量。H1299细胞分别转染Flag(A组)、Flag-BAG3质粒(B组),A549细胞分别转染siControl(C组)和siBAG3(D组)。通过蛋白免疫印迹实验检测质粒和小干扰RNA转染效率;采用CCK-8和平板克隆实验检测BAG3对NSCLC细胞增殖能力的影响;采用细胞划痕和Transwell实验检测BAG3对NSCLC细胞迁移能力的影响。结果BAG 3 mRNA在6种肿瘤中的表达均高于正常组织(t=8.45~24.13,P<0.05)。各NSCLC细胞系中BAG3的表达水平均明显高于Beas-2B细胞(t=5.76~15.34,P<0.05)。相比于A组,B组细胞中BAG3蛋白的表达水平显著升高(t=6.01,P<0.05);相比于C组,D组细胞中BAG3的蛋白水平明显降低(t=4.59,P<0.05);相比于A组,B组细胞增殖和迁移能力明显增强(t=5.12~17.10,P<0.05);相比于C组,D组细胞增殖和迁移能力明显下降(t=6.26~14.13,P<0.05)。结论BAG 3在NSCLC组织和细胞中高表达,并可促进NSCLC细胞的增殖和迁移。Objective To investigate the effect of Bcl-2-associated athanogene 3(BAG3)on the proliferation and migration abilities of non-small cell lung cancer(NSCLC)cells.Methods TCGA database was searched to perform a pan-cancer ana-lysis of BAG 3 expression in different tumors.Western blotting was used to measure the relative expression level of BAG3 protein in NSCLC cell lines(H1299,A549,H460,Anip973,PC9,H358,and 95-D cells)and the human normal lung epithelial cell line Beas-2B.H1299 cells were transfected with Flag(group A)or Flag-BAG3 plasmid(group B),and A549 cells were transfected with siControl(group C)or siBAG3(group D).Western blot was used to measure the transfection efficiency of plasmid and small interfering RNAs;CCK-8 assay and plate colony formation assay were used to observe the effect of BAG3 on the proliferation ability of NSCLC cells;cell scratch assay and Transwell assay were used to observe the effect of BAG3 on the migration ability of NSCLC cells.Results The mRNA expression level of BAG 3 in six types of tumor was significantly higher than that in normal tissue(t=8.45-24.13,P<0.05),and the expression level of BAG3 in each NSCLC cell line was significantly higher than that in the Beas-2B cell line(t=5.76-15.34,P<0.05).Compared with group A,group B had a significant increase in the protein expression level of BAG3(t=6.01,P<0.05),and compared with group C,group D had a significant reduction in the protein expression level of BAG3(t=4.59,P<0.05).Compared with group A,group B had significant increases in the proliferation and migration abilities of cells(t=5.12-17.10,P<0.05),and compared with group C,group D had significant reductions in the proliferation and migration abilities of cells(t=6.26-14.13,P<0.05).Conclusion There is a significantly high expression level of BAG 3 in NSCLC tissue and cells,which significantly promotes the proliferation and migration of NSCLC cells.
