摘  要:[目的]探索一种以鸡蛋蛋黄为原料,利用HGMS6菌株生物转化生产4-雄烯二酮(AD)的工艺,实现转化率60.78%。[方法]基于现有提取、转化技术,通过单因素和正交实验调整蛋黄胆固醇提取条件和分枝杆菌发酵条件,利用薄层析法和高效液相色谱法分析数据,提高AD的产率。[结果]明确最佳蛋黄胆固醇提取条件为:β-CD∶胆固醇=5∶1,水∶β-CD=15∶1,反应温度为55℃。提取率为98.12%。较吐温-80,添加环糊精时,得到的胆固醇转化率(60.18%)更高。[Objective]To explored a method of using egg yolk as the raw material,and biotransforming 4-androstenedione(AD)by HGMS6 strain,achieving a conversion rate of 60.78%.[Method]Based on the existing extraction and transformation technologies,the extraction conditions of egg yolk cholesterol and the fermentation environment of actinomycetes were adjusted,and the data were analyzed by thin-layer chromatography and high-performance liquid chromatography to improve the yield of AD.[Result]The optimal extraction conditions of egg yolk cholesterol were determined as follows:β-CD:cholesterol=5:1,water:β-CD=15:1,reaction temperature was 55 C.The extraction rate was 98.12%.Compared with Tween-80,adding cyclodextrin resulted in a higher cholesterol conversion rate(60.18%).
