摘 要:胰岛淀粉样多肽(islet amyloid polypeptide,IAPP)与胰岛素由β细胞共同分泌,当胰岛素抵抗时2种蛋白均会增多,β细胞因加工超载积累错误折叠的IAPP,损伤胰岛正常分泌功能。然而,IAPP还可产生活性氧、激活多种信号通路、干扰细胞代谢、在细胞内形成沉淀物、与其他淀粉样蛋白交叉折叠等方式,导致不同程度的细胞或组织损伤,最终引起全身多系统疾病。然而,IAPP在其他系统疾病发生发展过程中发挥的作用尚未形成广泛共识,有关研究结果也未被有效整合。本文将系统、全面地阐述不同结构形态的IAPP对神经、循环、运动、免疫等系统疾病的作用机制,有助于探索IAPP相关系统疾病的病理机制,为制定治疗和预防策略提供新思路。Islet amyloid polypeptide(IAPP)is co-secreted with insulin byβ-cells.When insulin resistance occurs,both proteins are elevated,leading toβ-cell damage due to the accumulation of misfolded IAPP during processing overload.Studies indicate that IAPP also generates reactive oxygen species,activates various signaling pathways,interferes with cell metabolism,form intracellular aggregates,and cross-fold with other amyloid proteins.These actions can lead to varying degrees of cell or tissue damage,resulting in multi-system diseases.However,the role of IAPP in the development of other system diseases has not reached a broad consensus,and the relevant research findings haven′t been effectively integrated.This article aims to comprehensively elaborate the impact of IAPP in different structural forms on the nervous,circulatory,motor,and immune system diseases,offering insights into the pathological mechanisms as well as new treatment and prevention strategies.
