摘  要:目的:探讨血必净注射液对肺缺血再灌注损伤的保护作用及其相关机制。方法:选择18只健康成年雄性大鼠,随机分为正常对照组(Sham组)、肺缺血再灌注组(LIR组)、血必净注射液组(XBJ组),采用开胸夹闭左肺门构建肺缺血再灌注损伤大鼠模型,苏木素—伊红染色观察各组大鼠肺组织病理学改变,检测各组大鼠肺组织湿/干重比(W/D)及肺组织炎症因子IL-1β、TNF-α水平,Western Blot检测各组肺组织PTEN、PI3K、Akt、mTOR、LC3-Ⅰ、LC3-Ⅱ、p62蛋白表达,qRT-PCR检测各组肺组织PTEN、PI3K、Akt、mTOR mRNA表达。结果:血必净注射液可显著减轻肺缺血再灌注大鼠肺组织损伤,降低缺血再灌注大鼠肺组织W/D及IL-1β、TNF-α含量(P<0.05)。与Sham组相比,LIR组、XBJ组PTEN、PI3K、Akt、mTOR、LC3-Ⅰ、LC3-Ⅱ、p62表达均升高(P<0.05);与LIR组相比,XBJ组PTEN、LC3-Ⅰ、LC3-Ⅱ、p62表达降低(P<0.05),PI3K、Akt、mTOR表达升高(P<0.05)。与Sham组相比,LIR组、XBJ组PTEN、PI3K、Akt、mTOR mRNA表达升高(P<0.05);与LIR组相比,XBJ组PTEN mRNA表达降低(P<0.05),PI3K、Akt、mTOR mRNA表达升高(P<0.05)。结论:血必净注射液可能通过抑制PTEN,激活PI3K/Akt/mTOR信号通路抑制肺缺血再灌注损伤自噬水平,从而改善肺缺血再灌注损伤,达到保护肺组织的作用。Objective:To investigate the protective effect of Xuebijing Injection on lung ischemia-reperfusion injury and its related mechanism.Methods:Eighteen healthy adult male rats were randomly divided into normal control group(sham group),lung ischemia-reperfusion group(LIR group)and Xuebijing injection group(XBJ group).The rat model of lung ischemia-reperfusion injury was established by clamping the left hilum of lung with thoracotomy.The pathological changes of lung tissue were observed by he staining,and the wet/dry weight ratio(W/D)and inflammatory factor IL-1β,TNF-α in lung tissue were detected.The protein expressions of PTEN,PI3K,Akt,mTOR,LC3-Ⅰ,LC3-Ⅱ,and p62 were detected by Western Blot,and the mRNA expressions of PTEN,PI3K,Akt,and mTOR were detected by qRT-PCR.Results:Xuebijing injection can significantly reduce lung tissue injury,W/D,and IL-1β,TNF-α content in rats with lung ischemia-reperfusion injury(P<0.05).Compared with Sham group,the expressions of PTEN,PI3K,Akt,mTOR,LC3-Ⅰ,LC3-Ⅱ,and p62 were increased in LIR group and XBJ group(P<0.05).Compared with LIR group,the expression of PTEN,LC3-Ⅰ,LC3-Ⅱ,p62 in XBJ group was decreased(P<0.05),the expression of PI3K,Akt and mTOR were increased(P<0.05).Compared with Sham group,the mRNA expressions of PTEN,PI3K,Akt,and mTOR in LIR group and XBJ group were increased(P<0.05).Compared with LIR group,the expression of PTEN mRNA in XBJ group was decreased(P<0.05),and the expression of PI3K,Akt,mTOR mRNA were increased(P<0.05).Conclusion:Xuebijing injection may inhibit autophagy in lung ischemia-reperfusion injury by inhibiting PTEN and activating PI3K/Akt/mTOR signaling pathway,so as to improve lung ischemia-reperfusion injury and protect lung tissue.
