摘 要:烟酰胺单核苷酸广泛应用在食品、制药、美容等领域。它是NAD+的关键前体之一,其具有很大的抗衰老的潜力,受到科学界和工业界的广泛关注。本研究首先构建出表达烟酰胺核糖激酶的重组E.coli BL21(DE3)/pET28b-NRK,然后优化蛋白表达条件,再通过单因素实验优化酶催化反应条件。结果表明最佳催化反应条件为:30℃,pH 7.0,1 g/L粗酶液,6 g/L烟酰胺核糖,6 g/L ATP,4 g/L MgSO_(4),在10 mL的反应体系中振荡反应30 min,产物NMN可达6.384 g/L,收率为97.9%。本研究为酶法合成NMN的应用研究奠定了基础。Nicotinamide mononucleotide was widely used in food,pharmaceutical,beauty and other fields.It was one of the key precursors of NAD^(+),which has great anti-aging potential,and has been widely concerned by the scientific and industrial communities.Recombinant E.coli BL21(DE3)/pET28b-NRK expressing nicotinamide ribokinase was constructed,and then the expression conditions of the protein were optimized,and the enzymatic reaction conditions were optimized by single factor experiment.The results showed that the optimal catalytic reaction conditions were as follows:at 30℃,pH 7.0,1 g/L crude enzyme solution,6 g/L nicotinamide ribose,6 g/L ATP,4 g/L MgSO_(4),the product NMN reached 6.384 g/L and the yield was 97.9% after incubation for 30 min in a 10 mL reaction system.This study laid a foundation for the application of enzymatic synthesis of NMN.
