摘 要:目的:制备Fe_(3)O_(4)/N-异丙基丙烯酰胺(PNIPAM)新型穴位敷贴剂型,检测其性能,并应用于哮喘模型的穴位贴敷疗法,探讨该疗法对哮喘大鼠辅助性T细胞2细胞因子调节作用。方法:制备Fe_(3)O_(4)/PNIPAM复合水凝胶并测定升温性能,电镜下观察内部结构,检验细胞毒性。将32只SD大鼠随机分为4组,分别为正常组、模型组、激光姜汁组、激素组,每组8只。除正常组外,其余组制备哮喘模型,激光姜汁组姜汁水凝胶贴敷大椎穴5 min并808 nm激光照射控制药物释放;激素组采用地塞米松1.2 mg/kg腹腔注射;均为隔日1次。观察大鼠一般状态;ELESA检测支气管肺泡灌洗液(BALF)中免疫球蛋白E(IgE)、白介素(IL)-4、IL-13的表达;HE染色检测肺组织病理情况。结果:2.0 Fe_(3)O_(4)/PNIPAM具有疏松孔状结构,良好的吸热升温性能,且无细胞毒性。与模型组比较,激光姜汁组可显著改善哮喘大鼠的行为学评分(P<0.01);与模型组和激素组比较,激光姜汁组显著降低BALF中IgE的含量,降低IL-4、IL-13的表达(P<0.01,P<0.05)。结论:Fe_(3)O_(4)/PNIPAM无细胞毒性,具有良好的疏松孔状结构可负载药物,具有良好的吸热升温性能可控制药物释放,可作为新型穴位贴敷制剂。姜汁水凝胶穴位贴敷疗法可通过降低BALF中IgE、IL-4、IL-13的表达缓解哮喘的发作症状。Objective:To prepare a new type of acupoint application form of Fe_(3)O_(4)/PNIPAM,test its performance,and apply it to acupoint application therapy in asthma animal model,and explore the effect of this therapy on the regulation of helper T cell 2 cytokines in asthmatic rats.Methods:Fe_(3)O_(4)/PNIPAM composite hydrogel was prepared and the warming performance was determined,and the internal structure was observed under the electron microscope to test the cytotoxicity.Thirty-two SD rats were randomly divided into 4 groups,which were the normal group,the model group,the laser-ginger juice group,and the hormone group,8 in each group.Except for the normal group,asthma models were prepared for the other groups.In the laserginger juice group,ginger juice hydrogel was applied to the Dazhui (DU14) for 5 min and 808 nm laser irradiation was used to control drug release;in the hormone group,dexamethasone was injected intraperitoneally at 1.2 mg/kg,and was administered once every other day.The general state of the rats was observed;the expression of IgE,IL-4 and IL-13 in BALF was detected by ELESA;and the pathology of lung tissue was detected by HE staining.Results:2.0 Fe_(3)O_(4)/PNIPAM had a loose pore-like structure,good heat-absorbing and warming properties,and no cytotoxicity.Compared with the model group,the laser-ginger juice group significantly improved the behavioral scores (P<0.01).Compared with the model group or the hormone group,the laser-ginger juice group significantly reduced the content of IgE,and the expression of IL-4 and IL-13 in the BALF of the asthmatic rats (P<0.01,P<0.05).Conclusion:Fe_(3)O_(4)/PNIPAM is non-cytotoxic,has a good loose pore structure for drug loading,and has good heat-absorbing and warming properties for controlling drug release,and can be used as a new type of acupoint application preparation.Ginger juice hydrogel acupoint application therapy can alleviate asthma attack symptoms by reducing the expression of IgE,IL-4,and IL-13 in BALF.
