摘 要:该研究通过分子模拟技术优化筛选玉米赤霉烯酮(zearalenone,ZEN)特异性多肽,并建立快速监测体系,预防ZEN中毒。分别用异硫氰酸荧光素和量子点标记模拟肽基于荧光共振能量转移的原理建立ZEN快速检测体系,基于荧光共振能量转移异硫氰酸检测方法,ZEN浓度0.1~40.0μg/L,线性关系(R^(2)=0.9907),检出限(S/N=3)为0.04μg/L。基于荧光共振能量转移量子点检测方法,ZEN浓度对数在0.08~65.00μg/L,线性关系(R^(2)=0.9929),检出限为0.03μg/L。对两种方法进行的特异性和稳定性评价显示出良好的结果。The molecular simulation technique was employed to optimize the detection method of the specific polypeptides of zearalenone(ZEN).A rapid detection system was established to prevent the poisoning caused by ZEN.The ZEN rapid detection system was established based on the principle of fluorescence resonance en⁃ergy transfer by using fluorescein isothiocyanate and quantum dot⁃labeled simulated peptides,respectively.The method established based on fluorescence resonance energy transfer with isothiocyanate labeling showed good linear relationship(R^(2)=0.9907)within the ZEN concentration of 0.1⁃40.0μg/L and limit of detection(S/N=3)being 0.04μg/L.The method established based on the fluorescence resonance energy transfer with quantum dot labeling showed good linear relationship(R^(2)=0.9929)within the ZEN concentration of 0.08⁃65.00μg/L and the limit of detection being 0.03μg/L.Both methods demonstrated high specificity and stability.
