摘  要:该研究以人神经母细胞瘤细胞(SH-SY5Y)探讨美藤果油对Aβ_(25-35)诱导阿尔兹海默症模型细胞的潜在保护作用。经Aβ_(25-35)和不同剂量的美藤果油干预后,测定美藤果油对AD模型细胞的抗氧化、抗凋亡等指标的变化。结果显示,在0~40μg/mL的剂量下,美藤果油对细胞活力不会产生影响,在0~100μg/L的剂量下,Aβ_(25-35)对细胞损伤程度上升,选用25μg/L的剂量诱导AD细胞模型。培养完成后,测定美藤果油对细胞的SOD、MDA水平,同时利用流式细胞术分析细胞凋亡程度、测定线粒体膜电位以及ROS水平,并采用western blot技术研究细胞Nrf-2、HO-1、Bcl-2、Bax蛋白表达水平。结果显示,20、40μg/mL剂量组细胞MDA含量水平分别降低了24.24%、42.73%,ROS含量水平分别降低了15.34%、25.53%,SOD的含量分别提高了20.81%、27.27%(P<0.005)。此外,美藤果油能够回调细胞线粒体膜电位,改善细胞凋亡,同时上调Nrf-2/HO-1抗氧化通路表达,增加抗凋亡蛋白Bcl-2表达、下调促凋亡蛋白Bax的表达。综上所述,美藤果油能够通过改善氧化应激损伤,逆转细胞凋亡来达到保护细胞的作用,其机制可能与增强Nrf-2/HO-1抗氧化通路,调节凋亡相关蛋白表达有关。Human neuroblastoma cells(SH-SY5Y)were utilized to investigate the potential protective effects of sacha inchi oil against Aβ_(25-35)-induced Alzheimer,s disease(AD)cells.Following exposure with Aβ_(25-35)and varying doses of sacha inchi oil,changes in antioxidant and anti-apoptotic indexes of the AD cell model caused by sacha inchi oil were measured.Results showed that sacha inchi oil,at doses ranging from 0 to 40μg/mL,did not affect cell viability.However,doses ranging from 0 to 100μg/L exacerbated Aβ_(25-35)-induced cell damage,with a 25μg/L dose selected to induce the AD cell model.Post-cultivation,the effects of sacha inchi oil on the superoxide dismutase(SOD)and malondialdehyde(MDA)levels were measured.Flow cytometry was employed to assess the degree of apoptosis,mitochondrial membrane potential,and reactive oxygen species(ROS)levels.Western blot analysis was performed to measure Nrf-2,HO-1,Bcl-2,and Bax protein expression levels.Results showed that supplementation with 20μg/mL and 40μg/mL of sacha inchi oil decreased MDA levels by 24.24%and 42.73%(P<0.005)and the levels of ROS by 15.34%and 25.53%(P<0.005),and increased levels of SOD by 20.81%and 27.27%(P<0.005),respectively.Furthermore,sacha inchi oil modulated mitochondrial membrane potential,attenuated apoptosis,upregulated Nrf-2/HO-1 antioxidant pathway expression,increased Bcl-2 anti-apoptotic protein expression,and down-regulated Bax pro-apoptotic protein expression.In conclusion,sacha inchi oil demonstrates cell protection by mitigating oxidative stress damage and reversing apoptosis.Its mechanism may involve enhancing the Nrf-2/HO-1 antioxidant pathway and regulating apoptosis-related protein expression.
