摘 要:该文以新吉细毛羊与小尾寒羊为研究对象。使用miRBase数据库、GO功能分析和KEGG通路富集分析等生物信息学方式对miR-485-3p、miR-154-3p/5p基因簇进行靶基因预测和分析。结果表明:miR-485-3p交集靶基因72个、miR-154-3p交集靶基因29个、miR-154-5p交集靶基因23个。GO功能分析与KEGG分析提示靶基因可与小蛋白偶联进行蛋白质修饰、和蛋白质泛素化以及细胞衰老、昼夜节律和NOD样受体通路相关;转录因子预测可知共同转录因子为235个;通过TF-miRNA-mRNA调控网络图可知,miR-485-3p、miR-154-3p和miR-154-5p基因簇上游转录因子和下游靶基因共同调控生物体过程;分析提示TRIM28、ZNF92和SPI1等转录因子在mRNA上游扩大miRNA基因簇强沉默功能,使oar-miR-485-3P、oar-miR-154-3p和oar-miR-154-5p通过靶基因DYRK1A参与细胞周期、细胞增殖和细胞凋亡从而影响羊毛纤维机制的表达。Xinji fine wool sheep and small-tailed Han sheep were used as the object of study.This study provided an idea for the regulatory mechanism of miR-485-3p and miR-154-3p/5p gene clusters in the plasma exosomes of Xinji fine wool sheep and small-tailed Han sheep on wool fiber.The bioinformatics methods such as miRBase database,GO function analysis and KEGG pathway enrichment analysis were used to predict and analyze the target genes of miR-485-3p and miR-154-3p/5p gene clusters.The results showed that there were 72 miR-485-3p intersection target genes,29 miR-154-3p intersection target genes,23 miR-154-5p intersection target genes.GO function analysis and KEGG pathway enrichment analysis suggested that target genes might be associated with small protein coupling for protein modification,protein ubiquitination,cell aging,circadian rhythm,and NOD-like receptor pathways.The number of common transcription factors was 235 by the prediction of transcription factors.Through the TF-miRNA-mRNA regulatory network diagram,the upstream transcription factors and downstream target genes of miR-485-3p,miR-154-3p and miR-154-5p gene clusters jointly regulated biological processes.The analysis suggested that transcription factors such as TRIM28,ZNF92 and SPII expanded the strong silencing function of miRNA gene clusters upstream of mRNA,so that oarmiR-485-3P,oar-miR-154-3p and oar-miR-154-5p were involved in cell cycle,cell proliferation and apoptosis through the target gene DYRK1A,thus affecting the expression of wool fiber mechanism.
