摘　　要：目的分析S100钙结合蛋白A6(S100A6)对EGFR 19号外显子缺失的人肺腺癌细胞吉非替尼耐药性的影响。方法本实验时间为2023年6—12月。取对数生长期EGFR 19号外显子缺失的人肺腺癌细胞PC9细胞和人正常肺上皮细胞BEAS-2B细胞,采用qRCR法检测其S100A6 mRNA表达水平,采用Western blotting法检测其S100A6表达水平。取对数生长期PC9细胞,将其分为实验组(转染S100A6基因过表达载体)和对照组(空PC9细胞)。采用MTT法检测两组不同浓度(0、0.1、1.0、10.0μmol/L)吉非替尼干预24、48、72 h时的细胞抑制率。结果PC9细胞中S100A6 mRNA及其蛋白表达水平低于BEAS-2B细胞(P<0.05)。0μmol/L吉非替尼干预24、48、72 h及10.0μmol/L吉非替尼干预24 h时,两组细胞抑制率比较,差异无统计学意义(P>0.05);0.1、1.0μmol/L吉非替尼干预24、48、72 h及10.0μmol/L吉非替尼干预48、72 h时,实验组细胞抑制率低于对照组(P<0.05)。结论过表达S100A6会促进EGFR 19号外显子缺失的人肺腺癌细胞对吉非替尼耐药。Objective To investigate the effect of S100 calcium binding protein A6(S100A6)on the gefitinib resistance of human lung adenocarcinoma cells with EGFR exon 19 deletion.Methods This study was performed from June to December 2023.Human lung adenocarcinoma cells with EGFR exon 19 deletion PC9 cells and human normal lung epithelial cells BEAS-2B cells at logarithmic growth stage were taken.The mRNA expression level of S100A6 in them was detected by qRCR method,and the S100A6 expression level in them was detected by Western blotting method.PC9 cells at logarithmic growth stage were divided into experimental group(transfected with S100A6 gene overexpression vector)and control group(empty PC9 cells).MTT method was used to detect the cell inhibition rates under the intervention of different concentrations(0,0.1,1.0,10.0μmol/L)of gefitinib at 24,48,and 72 hours in the two groups.Results The expression levels of S100A6 mRNA and protein in PC9 cells were lower than those in BEAS-2B cells(P<0.05).When treated with 0μmol/L gefitinib for 24,48,72 h and 10.0μmol/L gefitinib for 24 h,there was no significant difference in cell inhibition rate between the two groups(P>0.05).When treated with 0.1 and 1.0μmol/L gefitinib for 24,48,72 h and 10.0μmol/L gefitinib for 48,72 h,the cell inhibition rate of the experimental group was lower than that of the control group(P<0.05).Conclusion Overexpression of S100A6 can promote the resistance of human lung adenocarcinoma cells with EGFR exon 19 deletion to gefitinib.
