摘  要:目的研究白及多糖(BSP)对脂多糖(LPS)诱导小鼠急性肝损伤的作用与机制。方法将60只昆明雄性小鼠随机分为空白组(Control组)、模型组(LPS 1mg/kg,Model组)、阳性对照组(2mg/kg联苯双酯滴丸,BDP组)、BSP低、中、高剂量给药组(BSP 100、200、400mg/kg组),Control组和Model组给予生理盐水,其余4组按相应药物剂量,1次/d,连续灌胃7d,最后一次给药2h后,除Control组给予等体积生理盐水,其余5组腹腔注射LPS(1mg/kg)建立急性肝损伤模型,12h后取材,计算肝脏指数,测定血浆中AST、ALT、MDA、SOD、GSH-Px、CAT含量。光镜下观察HE染色后各组小鼠肝脏的组织变化,Western blot法检测各组小鼠肝组织中TLR4/MyD88/NF-κB和TNF-α/IL-1β/IL-6蛋白表达水平。结果与Control组相比,Model组肝脏指数明显上升(P<0.01),血浆中AST、ALT、MDA含量升高(P<0.01),SOD、GSH-Px、CAT含量显著下降(P<0.01),光镜下可见Model组肝小叶结构不清,伴有大量炎症细胞浸润;与Model组相比,BSP干预后降低了肝脏指数,增加了SOD、GSH-Px、CAT含量(P<0.01),显著降低血浆中AST、ALT、MDA含量(P<0.01),TLR4/MyD88/NF-κB和TNF-α/IL-1β/IL-6蛋白表达显著性下调(P<0.05)。结论BSP对LPS诱导的小鼠急性肝损伤具有保护作用,其机制可能与抗氧化及抑制TLR4/MyD88/NF-κB信号通路蛋白和减少TNF-α/IL-1β/IL-6等炎症因子的表达有关。Objective To study the effect and mechanism of Bletilla striata polysaccharide(BSP)on lipopolysaccharide(LPS)-induced acute liver injury in mice.Methods Sixty Kunming male mice were randomly divided into control group,model group(LPS 1mg/kg,Model group),positive control group(2mg/kg BifendatePills,BDP group),low,medium and high dose BSP administration groups(BSP 100,200,400mg/kg group).The control group and the model group were given normal saline,and the other four groups were given the corresponding drug dose once a day for 7 days.Two hours after the last administration,except for the control group,the other five groups were intraperitoneally injected with LPS(1mg/kg)to establish the model of acute liver injury.Twelve hours after the modeling,liver and plasma were corrected.Subsequently,the liver index was calculated,and the contents of AST,ALT,MDA,SOD,GSH-Px,and CAT in plasma were measured.The expression levels of TLR4/MyD88/NF-κB and TNF-α/IL-1β/IL-6 proteins in the liver tissue of mice in each group were detected by Western blotting.Results Compared with the control group,the liver index of the model group was significantly increased(P<0.01),the contents of AST,ALT and MDA in plasma were increased(P<0.01),and the contents of SOD,GSH-Px and CAT were significantly decreased(P<0.01).The structure of liver lobule was not clear in the model group accompanied by a large number of inflammatory cell infiltration.Compared with the model group,BSP intervention significantly reduced the liver index,increased the contents of SOD,GSH-Px and CAT(P<0.01),and significantly decreased the contents of AST,ALT and MDA in plasma(P<0.01).The protein expressions of TLR4/MyD88/NF-κB and TNF-α/IL-1β/IL-6 were significantly down-regulated(P<0.05).Conclusion BSP has a protective effect on LPS-induced acute liver injury in mice,and its mechanisms may be related to antioxidant and inhibition of TLR4/MyD88/NF-κB signaling pathway proteins and reduction of the expression of inflammatory factors such as TNF-α/IL-1β/IL-6.
