摘  要:为探究高能电子束辐照对藏成药石榴健胃散品质的影响,分别采用0、3、5、7、10、15 kGy剂量的电子束辐照处理石榴健胃散,探究辐照对其微生物含量、理化指标和有效活性成分含量的影响,并采用顶空固相微萃取-气相色谱-质谱联用技术鉴定挥发性成分。结果表明,电子束辐照能有效降低石榴健胃散中需氧菌、霉菌和酵母菌总数,且辐照剂量越大,微生物存活数越低。辐照剂量为7 kGy以上时,样品中的微生物降低至《中华人民共和国药典2020年版一部》的要求限度。辐照前后在所有样品中共鉴定出66种挥发性成分,包括6种醛类、10种醇类、4种酮类、26种烯烃类、7种烷烃类、3种酸类、4种酯类、5种杂环类及芳香类、1种醚类。7 kGy以下的辐照剂量对主要挥发性成分的影响较小且含量稳定。经不同辐照剂量处理后石榴健胃散指纹图谱的相似度均为1.000,表明辐照前后石榴健胃散整体成分较为稳定,主要活性成分含量无显著差异。综上,7 kGy辐照剂量能达到良好的灭菌效果,可保障石榴健胃散的品质。本研究结果可为电子束辐照技术在石榴健胃散贮藏中的应用提供参考依据。To investigate the effect of high-energy electron beam irradiation sterilization on the quality of Shiliu Jian-Wei Powder in Tibetan medicine,samples were treated with electron beam irradiation at doses of 0,3,5,7,10,and 15 kGy,respectively.The effects of irradiation on the microbial content,physicochemical properties,and the content of the effective active ingredients were investigated.Volatile components were identified by headspace solid-phase microextraction-gas chromatography-mass spectrometry.The results showed that electron beam irradiation could effectively reduce the total number of bacteria,molds,and fungal yeasts in Shiliu Jian-Wei Powder.With the increasing of irradiation doses,the number of microorganisms reduced.At irradiation doses of over 7 kGy,the microbial counts in Shiliu Jian-Wei Powder were reduced to the required limit based on the Pharmacopoeia standards.A total of 66 volatile components were identified in all samples before and after irradiation,including 6 aldehydes,10 alcohols,4 ketones,26 olefins,7 alkanes,3 acids,4 esters,5 heterocyclic and aromatic compounds,and 1 ether.Irradiation doses up to 7 kGy had little effect on the main volatile components.The similarity of fingerprints of Shiliu Jian-Wei Powder with different irradiation doses of the electron beam was 1.000.This indicated that the overall components were stable before and after irradiation,and no significant difference was found in the content of the main active ingredients.In conclusion,the irradiation dose of 7 kGy can achieve good sterilization effect,which can ensure the quality of Shiliu Jian-Wei Powder.The findings could provide a certain reference for the application of electron beam irradiation technology in the storage of Shiliu Jian-Wei Powder.
