摘 要:研究山莴苣素通过调控线粒体功能对游离脂肪酸(FFA)诱导HepG2细胞脂质堆积的影响,探索抗非酒精性脂肪肝病(NAFLD)机制。FFA诱导HepG2细胞脂质变性(构建NAFLD体外模型),给予山莴苣素共同处理48 h。采用油红O染色观察细胞内脂滴含量,检测细胞内甘油三酯(TG)含量,脂肪酸β氧化的活力以及高通量电子显微镜观察MitoSOX Red与Mitotracker Red的荧光强弱;qRT-PCR法检测脂质代谢与线粒体相关基因的相对表达量;蛋白质印迹法测定PPARα的蛋白水平。结果显示,与模型组相比,山莴苣素抑制FFA诱导HepG2细胞内脂滴的增加,降低TG含量、MitoSOX Red的荧光强度以及SREBP1的mRNA相对表达量;上调脂肪酸β氧化的活力、Mitotracker Red的荧光强度以及AMPK、PPARα、CPT1A、PPARγ、PINK1、Parkin基因的相对表达量和PPARα蛋白表达水平。山莴苣素通过PPARα/CPT1A,PINK1/Parkin的信号转导恢复线粒体功能,改善FFA诱导HepG2细胞脂质堆积。Investigating the effect of Lactucin on free fatty acid(FFA)-induced lipid accumulation in HepG2 cells through the regulation of mitochondrial function,to explore the mechanism against non-alcoholic fatty liver disease(NAFLD).HepG2 cells were induced with FFA to induce lipid accumulation(establishing an NAFLD in vitro model)and treated with Lactucin for 48 hours.Oil red O staining was used to observe the content of intracellular lipid droplets,detect intracellular triglyceride(TG)content,assess the fatty acidβ-oxidation activity,and observe the fluorescence intensity of MitoSOX Red and Mitotracker Red using high throughput electron microscopy.the relative expression levels of lipid metabolism and mitochondrial-related genes were measured by qRT-PCR,and Western blotting was conducted to determine the protein levels of PPARα.The results showed that compared to the model group,Lactucin inhibited the increase of intracellular lipid droplets induced by FFA in HepG2 cells,reduced TG content,fluorescence intensity of MitoSOX Red,and relative mRNA expression of SREBP1.It also up-regulation fatty acidβ-oxidation activity,the fluorescence intensity of Mitotracker Red,as well as the relative expression levels of AMPK,PPARα,CPT1A,PPARγ,PINK1,Parkin genes,and the protein expression level of PPARα.Lactucin restored mitochondrial function through the PPARα/CPT1A and PINK1/Parkin signaling pathways,improving lipid accumulation induced by FFA in HepG2 cells.
