摘 要:目的:探讨黄精多糖(PSP)对口腔溃疡模型大鼠的治疗效果及作用机制。方法:将60只大鼠随机分为正常组、模型组和PSP低(50mg/kg)、中(100mg/kg)、高(200mg/kg)剂量组,除正常组外均采用机械创伤联合氢氧化钠(NaOH)晶体灼烧建立口腔溃疡模型。造模后连续灌胃给药10d,正常组和模型组大鼠给予生理盐水,PSP低、中、高剂量组大鼠接受相应浓度的PSP溶液。在给药前以及给药第3、5、7、10d,测量各组大鼠口腔溃疡的最大横径和最大纵径,计算溃疡面积。同时通过酶联免疫吸附试验(ELISA)检测大鼠血清中肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)水平,采用水溶性四唑蓝法(WST)和硫代巴比妥酸法(TBA)检测大鼠血清中超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果:与模型组比较,PSP药物组大鼠的溃疡面积缩小更为显著,尤其是PSP中、高剂量组(P<0.05)。PSP低、中、高剂量组能显著降低大鼠造模后血清中升高的TNF-α、IL-6及MDA含量(P<0.05),同时增加血清SOD水平(P<0.05)。结论:PSP对机械化学联合法诱导的口腔溃疡模型大鼠有明显的治疗作用,能促进溃疡病变的愈合,其机制可能与其抗炎、抗氧化作用有关。Objective:To investigate the therapeutic effects of Polygonatum sibiricum polysaccharides(PSP)on oral ulcer model rats and its mechanism.Methods:A total of 60 rats were randomly divided into the normal group,model group and PSP low-dose(50 mg/kg),medium-dose(100 mg/kg),and high-dose(200 mg/kg)groups.Except for the normal group,all the rats were mechanically traumatized and burned with sodium hydroxide(NaOH)crystals to establish oral ulcer models.After modeling,the drugs were administered by continuous gavage for 10 days.Normal saline was administered to the normal and model groups,while the low,medium,and high dose PSP groups were respectively administered with corresponding concentrations of PSP solution.The maximum transverse and longitudinal diameters of oral ulcers were measured before administration and on the 3rd,5th,7th and 10th days of administration in each group,and the area of ulcers was calculated.The levels of tumor necrosis factor(TNF)-αand interleukin(IL)-6 were measured using enzymelinked immunosorbent assay(ELISA),and the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)were measured using the water-soluble tetrazolium blue method(WST)and the thiobarbituric acid method(TBA),respectively,in the serum of rats.Results:The reduction of ulcer area was more significant in the PSP drug group,especially in the PSP medium-and high-dose groups,compared to the model group(P<0.05).The PSP low-,medium-,and highdose groups significantly reduced the elevated serum levels of TNF-α,IL-6,and MDA in the modeled rats(P<0.05),as well as increased the serum SOD level(P<0.05).Conclusion:PSP exhibited a significant therapeutic effect on oral ulcer model rats induced by mechanochemical chemical association method,which could significantly promote the healing of oral ulcer lesions.The treatment mechanism may be related to its anti-inflammatory and antioxidant effects.
