摘 要:试验旨在探究马尾藻多酚对大肠杆菌(Escherichia coli,E.coli)的抑菌活性。采用乙醇浸提法和葡聚糖凝胶柱层析法对马尾藻中的褐藻多酚进行提取纯化,通过测定褐藻多酚对E.coli的抑菌水平、最小抑菌浓度、最小杀菌浓度,以及对其增殖曲线、生物膜形成的影响,研究褐藻多酚对E.coli的抑制效果。结果表明:提取纯化的多酚含量为848mg/g,其对E.coli的最小抑菌浓度(MIC)为4 mg/mL,最小杀菌浓度(MBC)为16 mg/mL;与对照组相比,经2 MIC、1 MIC褐藻多酚处理的大肠杆菌生长明显受到抑制,1/2 MIC褐藻多酚处理组抑菌效果降低,4 MIC褐藻多酚处理组完全抑制了E.coli的生长,其中添加褐藻多酚的各组在2~8 h内均能极显著抑制细菌增殖(P<0.01);褐藻多酚浓度为4~16 mg/mL时可以显著抑制生物膜的形成(P<0.05)。综上,褐藻多酚对E.coli有明显的抑制作用。The experiment aims to investigate antibacterial activity of Sargassum polyphenols on Escherichia coli(E.coli).Ethanol extraction and dextran gel column chromatography were used to extract and purify phlorotannins from Sargassum,the inhibitory effect of phlorotannins on E.coli was investigated by measuring the inhibition level,minimum inhibitory concentration and minimum bactericidal concentration of phlorotannins on E.coli,as well as the influence on its proliferation curve and biofilm formation.The results show that the content of extracted and purified polyphenols was 848 mg/g.The minimum inhibitory concentration(MIC) for E.coli was 4 mg/mL and the minimum bactericidal concentration(MBC)was 16 mg/mL.Compared to the control group,the growth of E.coli treated with the 2 MIC and 1 MIC phlorotannins was significantly inhibited.The antibacterial effect of 1/2 MIC phlorotannins was reduced,and E.coli growth was completely inhibited in 4 MIC phlorotannins.All groups supplemented with phlorotannins had a significant inhibition of bacterial proliferation within 2 to 8 hours(P<0.01).Concentrations of phlorotannins in the range of 4 to 16 mg/mL were able to significantly reduce biofilm formation(P<0.05).In conclusion,the phlorotannins have a clear inhibitory effect on E.coli.
