摘  要:目的:探讨研究虎杖苷调控Janus激酶1(JAK1)/信号传导和转录激活因子3(STAT3)信号通路对小鼠宫颈癌细胞凋亡及裸鼠移植瘤的作用。方法:选取人宫颈癌HeLa细胞株,分为对照组(未经任何处理的HeLa细胞)、虎杖苷低、中、高剂量组(加入50、100、150μmol/L虎杖苷),继续培养24、48、72 h, Annexin V双染法检测HeLa细胞凋亡,MTT检测HeLa细胞增殖能力,Transwell小室检测HeLa细胞侵袭能力,qRT-PCR检测JAK1、STAT3 mRNA表达,Western blot检测JAK1/STAT3信号通路相关蛋白表达,并进行细胞形态学观察。结果:与对照组比较,虎杖苷低、中、高剂量组HeLa细胞凋亡率升高,高剂量组凋亡率高于中剂量组,中剂量组高于低剂量组(均P<0.05)。虎杖苷低、中、高剂量组HeLa细胞增殖和侵袭能力显著下降,虎杖苷高剂量组增殖率和侵袭细胞数最低(均P<0.05)。虎杖苷低、中、高剂量组HeLa细胞中JAK1、STAT3 mRNA和蛋白表达显著下降,高剂量组JAK1、STAT3 mRNA和蛋白表达低于中剂量组,中剂量组低于低剂量组(均P<0.05)。虎杖苷低、中、高剂量组小鼠肿瘤体积缩小,肿瘤质量降低(均P<0.05)。结论:虎杖苷可诱导宫颈癌HeLa细胞凋亡,抑制其增殖、迁移、侵袭及裸鼠皮下成瘤,其作用机制可能与调控JAK1/STAT3通路信号通路有关。Objective:To investigate the effect of polydatin on apoptosis of cervical cancer cells and transplanted tumor mice by regulating Janus kinase 1(JAK1)/signaling and transcriptional activator 3(STAT3)signaling pathway.Methods:Human cervical cancer HeLa cell lines were selected and divided into control group(HeLa cells without any treatment)and polydatin low,medium and high dose groups(50,100 and 150μmoL/L polydatin were added).The culture was continued for 24,48 and 72 hours.Annexin V double staining was used to detect the apoptosis of HeLa cells.The proliferation ability of HeLa cells was detected by MTT,the invasion ability of HeLa cells was detected by Transwell chamber,the mRNA expression of JAK1 and STAT3 was detected by qRT-PCR,and the expression of JAK1/STAT3 signaling pathway related proteins was detected by Western blot,and the cell morphology was observed.Results:Compared with the control group,the apoptosis rate of HeLa cells in polydatin low,medium and high dose groups was increased,the apoptosis rate in high dose group was higher than that in medium dose group,and that in medium dose group was higher than that in low dose group(all P<0.05).The proliferation and invasion ability of HeLa cells in low,medium and high doses of polydatin were significantly decreased,and the proliferation rate and invasion number of HELA cells in high dose group were the lowest(all P<0.05).The expressions of JAK1 and STAT3 mRNA and protein in HeLa cells of polydatin low,medium and high dose groups were significantly decreased,and the expressions of JAK1 and STAT3 mRNA and protein in high dose group were lower than those in medium dose group,and those in medium dose group were lower than those in low dose group(all P<0.05).The tumor volume decreased and the tumor quality decreased in the low,medium and high dose groups of polydatin(P<0.05).Conclusion:Polydatin can induce apoptosis of HeLa cells of cervical cancer,inhibit proliferation,migration,invasion and subcutaneous tumor formation in nude mice.The mechanism of action ma
