摘 要:目的以益生菌为对照组,分析骨化三醇对帕金森病(PD)小鼠模型黑质、结肠迷走神经α突触核蛋白的影响。方法选取雄性SPF级健康C57BL/6小鼠45只,随机分为A组(正常对照,n=5)、B1组(PD模型对照,n=10)、B2组(PD模型益生菌干预,n=10)、B3组(PD模型骨化三醇干预,n=10),B4组(PD模型益生菌联合骨化三醇干预,n=10)。每组均连续喂养3个月。喂养结束完成行为学、相关血液学检测后处死实验小鼠取各组n=3,分别取大脑黑质和结肠迷走神经组织,行Western blot检测a-突触核蛋白(a-syn)含量。结果结肠迷走神经中a-syn含量显示:(1)PD模型组(B1组)、益生菌干预组(B2组)、骨化三醇干预组(B3组)明显高于正常组(A组),^(a)P<0.01;(2)益生菌干预组(B2组)、骨化三醇干预组(B3组)、益生菌+骨化三醇联合干预组(B4组)均低于PD模型组(B1组),^(b)P<0.01;(3)骨化三醇干预组(B3组)高于益生菌干预组(B2组),^(c)P<0.01;(4)益生菌+骨化三醇联合干预组(B4组)低于骨化三醇干预组(B3组),^(d)P<0.01;黑质中的a-syn含量显示:(1)PD模型组(B1组)、益生菌干预组(B2组)明显高于正常组(A组),^(a)P<0.01;(2)益生菌干预组(B2组)、骨化三醇干预组(B3组)、益生菌+骨化三醇联合干预(B4组)均低于PD模型组(B1组),^(b)P<0.01;(3)骨化三醇干预组(B3组)、益生菌+骨化三醇联合干预(B4组)均低于益生菌干预组(B2组),^(d)P<0.01。结论(1)益生菌、骨化三醇对于PD小鼠肠道迷走神经中的a-syn的沉积均有抑制作用,而益生菌在此过程中显示更显著的作用;(2)益生菌、骨化三醇可不同程度减少PD小鼠黑质中的a-syn的沉积,其中骨化三醇对于α-syn中枢沉积的干预作用尤显著。Objective To analyze the effects of calcitriol on substantia nigra andαsynaptosynuclein(a-syn)of vagus nerve in PD mice model with probiotics as control group.Methods Forty-five male SPF healthy C57BL/6 mice were randomly divided into group A(normal control,n=5),group Bl(PD model control,n=10),group B2(PD model prohiotic intervention,n=10),and group B3(PD model calcitriol intervention,n=10).B4 group(PD model probiotics combined with calcitriol intervention,n=10).Each group was fed continuously for 3 months.After the completion of behavioral and related hematological tests,the experimental mice were killed and n=3 were taken from each group,and the substantia nigra of the brain and vagus nerve tissue of the colon were respectively taken for Western blot detection ofα-syn content.Results The content of a-syn in vagus nerve showed as follows:①PD model group(group B1),prohiotic intervention group(group B2)and calcitriol intervention group(group B3)were significantly higher than normal group(group A),^(a)P<0.01;②Probiotic intervention group(B2 group),calcitriol intervention group(B3 group)and probiotic+calcitriol combined intervention group(B4 group)were all lower than PD model group(B1 group),^(h)P<0.01.③Calcitriol intervention group(group B3)was higher than probiotic intervention group(group B2),^(c)P<0.01;4.The combination of prohiotics and calcitriol intervention group(B4 group)was lower than that of calcitriol intervention group(B3 group),^(d)P<0.01.The content of a-syn in substantia nigra showed as follows:①PD model group(B1)and prohiotic intervention group(B2)were significantly higher than normal group(A),^(a)P<0.01;②Probiotic intervention group(B2 group),calcitriol intervention group(B3 group),and prohiotic+calcitriol combined intervention group(B4 group)were all lower than PD model group(B1 group),^(b)P<0.01.③Calcitriol intervention group(group B3)and prohiotic+calcitriol combined intervention group(group B4)were lower than prohiotic intervention group(group B2),^(d)P<0.01.Conclusions①Both pro
