摘 要:目的总结蛋白质棕榈酰化的机制及与癌症发生发展的关系,通过分析棕榈酰化在消化系统肿瘤中的研究进展,探索靶向棕榈酰化过程用于消化系统肿瘤治疗方面的潜在策略.方法以"棕榈酰化、翻译后修饰、消化系统肿瘤、肿瘤治疗"为中文检索词,以"protein palmitoylation、post-translational modification、digestive system cancer、tumor therapy"为英文检索词,检索PubMed和中国知网数据库2007年1月-2023年7月的相关文献.纳入标准:(1)棕榈酰化的生化反应机制和特点;(2)棕榈酰化过程与癌症相关机制;(3)棕榈酰化在消化系统肿瘤中的作用;(4)靶向棕榈酰化途径对于肿瘤治疗的潜在作用.排除标准:(1)研究机制模糊文献;(2)会议性和评论类文献;(3)内容相似或重复文献.最终共纳入符合条件文献70篇.结果蛋白质棕榈酰化对于癌基因(如N-Ras和表皮生长因子受体)的功能至关重要.在调控蛋白质的转运、细胞定位和稳定性等方面都具有重要的作用,参与诸多生物学过程,并与消化系统肿瘤的发生发展关系密切.靶向棕榈酰化过程或联合肿瘤免疫治疗具有潜在治疗价值.结论棕榈酰化在消化系统肿瘤的发生发展及侵袭转移中具有重要地位,并影响肿瘤免疫.棕榈酰化研究有望为消化系统肿瘤的治疗提供新靶点.Objective To summarize the mechanism of protein palmitoylation and its correlation with the occurrence and de-velopment of cancer,elucidate the research progress of palmitoylation in digestive system cancer,and explore the poten-tial strategies of targeting palmitoylation for the treatment of digestive system cancer.Methods With"protein palmitoyla-tion,post-translational modification,digestive system cancer,tumor therapy"as the keywords,the relevant literature of PubMed and CNKI was retrieved from January 2007 to July 2023.Inclusion criteria:(1)Biochemical reaction mechanism and characteristics of palmitoylation;(2)The mechanism of palmitoylation process and cancer;(3)The role of palmitoyla-tion in digestive system cancer;(4)The potential role of targeting palmitoylation pathways in tumor treatment.Exclusion criteria:(1)Literature on the study of ambiguous mechanisms;(2)Conference,commentary and other literature;(3)Similar or duplicate literature.A total of 70 eligible literature were included.Results Protein palmitoylation is essen-tial for the function of oncogenes such as N-Ras and epidermal growth factor receptor.It plays an important role in regu-lating protein transport,cell localization and stability,participates in many biological processes,and is closely related to the occurrence and development of digestive system cancer.Targeting palmitoylation processes may be of potential thera-peutic value in combination with tumor immunotherapy.Conclusions Protein Palmitoylation plays an important role in the occurrence,development,invasion and metastasis of digestive system cancer,and affects tumor immunity.Palmitoylation research is expected to provide a new target for the treatment of digestive system cancer.
