摘 要:研究旨在探讨鸡骨草黄酮(ACE)能否减轻黄曲霉毒素B_(1)(AFB_(1))诱导的肝毒性作用,为临床防治AFB_(1)引起的肝损伤提供新思路。选择健康三黄鸡60羽,随机分为3组,每组4个重复,每个重复5只鸡。空白对照组(NC组)饲喂基础日粮,AFB_(1)组饲喂基础日粮+2.8 mg/kg AFB_(1),ACE组饲喂基础日粮+2.8 mg/kg AFB_(1)+1 g/kg ACE。试验期7 d。结果显示,与NC组相比,AFB_(1)组雏鸡14日龄体重、肝脏谷胱甘肽转移酶(GST)表达量以及肝脏过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性显著降低(P<0.05),雏鸡血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、碱性磷酸酶(ALP)和γ-谷氨酰转肽酶(GGT)活性及肝脏丙二醛(MDA)含量、肝脏细胞色素P450酶1A2 (CYP1A2)表达量显著升高(P<0.05)。与AFB_(1)组相比,ACE组雏鸡14日龄体重、肝脏GST表达量以及CAT、SOD、GSH-Px活性显著升高(P<0.05),雏鸡血清AST、ALT、ALP和GGT活性及肝脏MDA含量、肝脏CYP1A2表达量显著下降(P<0.05)。研究表明,日粮补充ACE可能通过增强抗氧化能力和抑制关键的细胞色素P450酶(CYP450)同工酶介导的AFB1对有毒产物AFB1-8,9-环氧化物(AFBO)的激活,起到协同作用,从而保护雏鸡免受AFB1诱导的肝损伤。The purpose of the study was to ACE could alleviate the hepatotoxicity induced by AFB_(1),and to provide a new idea for clinical prevention and treatment of liver injury induced by AFB_(1).60 healthy Sanhuang chickens were randomly divided into three groups,and four replicates in each group with five chickens in each replicate.Chickens in the blank control group(NC group)were fed basal diets,AFB_(1) group were fed basal diets plus 2.8 mg/kg AFB_(1),and ACE group were fed basal diets plus 2.8 mg/kg AFB_(1)+1 g/kg ACE.The experiment lasted for seven days.The results showed that compared with the NC group,the body weight,liver GST expression,liver CAT,SOD,and GSH-Px activities of the AFB1 group decreased significantly(P<0.05),serum AST,ALT,ALP and GGT activities,liver MDA content,and liver CYP1A2 expression increased significantly(P<0.05).Compared with the AFB1 group,the body weight,liver GST expression and activities of CAT,SOD and GSH-Px in the ACE group increased significantly(P<0.05),while serum AST,ALT,ALP and GGT activities,liver MDA content,and liver CYP1A2 expression in the ACE group decreased significantly(P<0.05).Studies have shown that dietary ACE supplementation may play a synergistic role by enhancing antioxidant capacity and inhibiting the activation of AFBO mediated by AFB1,which is a key CYP450 isozyme,thus protecting chickens from AFB_(1)-induced liver injury.
