摘  要:目的:评估归龙丸对高糖诱导的大鼠神经膜细胞(rats schwann cell,RSC96)分泌肿瘤坏死因子α(tumor necrosis factor α,TNF-α)、血清白细胞介素10(interleukin-10,IL-10)的影响。方法:将RSC96神经膜细胞复苏及培养,在含17 mmol/L的葡萄糖培养基中培养48 h。分为S1组(维生素D_(3)+等渗乙醇溶液)、S2组(维生素D_(3)+等渗乙醇溶液+归龙丸高剂量)、S3组(维生素D3+等渗乙醇溶液组+归龙丸中剂量)、S4组(维生素D_(3)+等渗乙醇溶液组+归龙丸低剂量)、S5组(维生素D_(3)+CYP24A1抑制剂genistein的乙醇溶液)等5组,S1~S4组中分别加入维生素D_(3)及与其等浓度的乙醇,S5组加入维生素D_(3)及CYP24A1抑制剂genistein的乙醇溶液,作用48 h。48 h后应用酶联免疫吸附试验法检测各组细胞上清液中TNF-α、IL-10的水平,并通过相应的基因引物检测RSC96细胞内CYP24A1m表达水平。结果:经归龙丸和CYP24A1抑制剂组处理后的神经膜细胞,其TNF-α的水平下降,IL-10水平上升,CYP24A1m表达水平下调。结论:归龙丸可能通过抑制CYP24A1蛋白表达,间接调节维生素D_(3)使神经膜细胞TNF-α分泌减少、IL-10分泌增加,从而对高糖诱导的RSC96神经膜细胞起到保护作用。Objective:To evaluate the effects of Guilong pills on the secretion of TNF-α and IL-10 in RSC96Schwann cells induced by high glucose.Methods:After being resuscitated and cultured in a medium containing 17 mmol/L glucose for 48 h,RSC96 Schwann cells were divided into five groups:the S1 group(Vitamin D_(3)+isotonic ethanol solution group),the S2 group(Vitamin D_(3)+isotonic ethanol solution+high dose group of Guilong pills),the S3 group(Vitamin D_(3)+isotonic ethanol solution+moderate dose group of Guilong pills),the S4 group(Vitamin D_(3)+isotonic ethanol solution+low dose group of Guilong pills) and the S5 group(Vitamin D_(3)+ethanol solution group of CYP24A1 inhibitor genistein),Vitamin D_(3) and the same concentration of ethanol solution were added respectively into the S1-S4 groups,ethanol solution of vitamin D_(3) and CYP24A1 inhibitor genistein was added to group S5 and acted for 48 hours.ELISA method was applied to detect the levels of TNF-α and IL-10 in the cell supernatant of each group in 48 hours,and the corresponding gene primer was used to measure the levels of CYP24A1m in RSC96 cells.Results:The levels of TNF-α decreased while the levels of IL-10 increased,the expressions of CYP24A1 downregulated in RSC96 Schwann cells processed with Guilong pills and CYP24A1m group.Conclusion:Guilong pills could reduce the secretion of TNF-α and increase the levels of IL-10 possibly by inhibiting the expressions of CYP24A1 protein,and indirectly regulating the levels of Vitamin D_(3),thereby protecting RSC96 Schwann cells induced by high glucose.
