摘 要:以苦木的根、茎、叶为试材,采用组织分离法从苦木的根、茎、叶中分离得到内生真菌,通过ITS序列分析,对其进行鉴定。采用K-B纸片法和最小抑制浓度法,研究了内生真菌发酵液对大肠杆菌和金黄色葡萄球菌的抑制效果,以期为苦木内生真菌在抗菌方面的研究价值提供参考依据,为寻找新型绿色高效的抗菌活性物质提供新思路。结果表明:苦木植物中蕴含丰富的内生真菌资源,从苦木根、枝、叶中分离得到16株内生真菌,分属于4门6纲8目8科8属,链格孢属(Ilternarsa)和葡萄座腔菌属(Botryosphaeria)为优势菌属。13株苦木内生真菌的发酵液提取物对2株指示菌表现出不同强度的抑菌作用,其中3株内生真菌的发酵液粗提物对大肠杆菌和金黄色葡萄球菌有较强抗菌活性,MIC值介于0.024~3.120μg·mL^(-1)。综合抑菌的广谱性和效能而言,G1菌株的抗菌效果最好。Taking the roots,stems,and leaves of Picrasma quassioides(D.Don)Benn.as experimental materials,endophytic fungi were isolated from the roots,stems,and leaves of Picrasma quassioides(D.Don)Benn.using tissue isolation method.ITS sequence analysis was used to identify them.The inhibitory effects of endophytic fungi fermentation broth on Escherichia coli and Staphylococcus aureus were studied using K-B paper method and minimum inhibitory concentration method,in order to provide reference for the research value of endophytic fungi in the antibacterial of Picrasma quassioides(D.Don)Benn.and to provide new ideas for finding new green and efficient antibacterial active substances.The results showed that Picrasma quassioides(D.Don)Benn.plants contain abundant endophytic fungal resources.16 endophytic fungi were isolated from the roots,branches,and leaves of Picrasma quassioides(D.Don)Benn.belonging to 4 phyla,6 classes,8 orders,8 families,and 8 genera.The dominant genera were Ilternarsa and Botryospaeria.The fermentation broth extracts of 13 endophytic fungi from Picrasma quassioides(D.Don)Benn.showed varying degrees of antibacterial activity against two indicator bacteria.Among them,the crude extracts of fermentation broth from 3 endophytic fungi showed strong antibacterial activity against Escherichia coli and Staphylococcus aureus,with MIC values ranging from 0.02 to 3.12μg·mL^(-1).In terms of comprehensive antibacterial broad-spectrum and efficacy,G1 strain has the best antibacterial effect.
