摘 要:目的 探讨在牙周炎微环境中低氧诱导因子-1α(HIF-1α)和H型血管是否参与牙周炎-牙槽骨吸收过程。方法 将10只C57BL/6小鼠随机分为牙周炎组和对照组,每组5只;牙周炎组采用5-0丝线结扎C57BL/6小鼠双侧上颌第二磨牙,建立小鼠实验性牙周炎模型;对照组不予处理。结扎14 d后micro-CT检测小鼠上颌第二磨牙牙槽骨高度的变化,免疫组化实验分析牙周炎牙槽骨缺损处组织蛋白酶K(CTSK)的表达情况,免疫荧光法分析牙周组织中H型血管(CD31+Emcn+)及HIF-1α的表达。结果 采用丝线结扎法成功构建小鼠牙周炎模型。micro-CT扫描结果示,与对照组相比,牙周炎组小鼠釉牙骨质界(CEJ)距牙槽嵴顶(ABC)的距离(CEJ-ABC)显著增加(P<0.001);HE染色结果显示,牙周炎组出现较重炎症病理性反应。免疫组化结果显示,牙周炎组牙槽骨中CTSK的表达水平相较对照组更高(P<0.05)。免疫荧光结果显示,与对照组相比,牙周炎组牙周组织中HIF-1α表达水平提高(P<0.000 1),H型血管生成增加,即CD31+Emcn+的表达更高(P<0.001)。结论 通过丝线结扎成功建立小鼠牙周炎模型,小鼠牙周炎牙周组织中HIF-1α和H型血管特异性升高,提示在牙周炎早期的缺氧微环境中,HIF-1α和H型血管参与牙周炎牙槽骨的骨吸收过程。Objective To investigate whether hypoxia inducible factor-1α(HIF-1α)and type H vessels are involved in the periodontitis microenvironment.Methods Ten C57BL/6 mice were randomly divided into periodontitis group and control group,with 5 mice in each group.The maxillary second molar of mice in periodontitis group were ligated with 5-0 silk thread to establish the experimental periodontitis model,while the mice in control group were not treated.After ligation for 14 d,the changes of alveolar bone height of the maxillary second molars were detected by micro-CT scanning,the expression of cathepsin K(CTSK)was analyzed by immunohistochemistry,and the formation of type H vessels(CD31+Emcn+)and the expression of HIF-1αin periodontal tissues were analyzed by immunofluorescence analysis.Results The experimental periodontitis model was successfully established by silk thread ligature.The micro-CT scanning results showed that the distance between the cementoenamel junctions and the alveolar bone crest(CEJ-ABC)of mice in periodontitis group was significantly higher than that in control group(P<0.001),and HE staining results demonstrated that the sevious inflammatory pathological reaction was observed in periodontitis group.Immunohistochemical analysis indicated that CTSK expression level was significantly higher in periodontitis group than in control group(P<0.05).Immunofluorescence analysis demonstrated that HIF-1αexpression level was increased in periodontitis group compared with control group(P<0.0001),and the number of type H vessels was also significantly increased in periodontitis group,as evidenced by higher densities of CD31+Emcn+(P<0.001).Conclusion The mouse periodontitis model is successfully established by silk ligation,and the expressions of HIF-1αand type H vessels are specifically elevated in the periodontal tissues within periodontitis,which suggests that HIF-1αand type H vessels may be involved in the process of bone resorption in periodontitis alveolar bone.
