摘 要:目的探讨胃肠活检组织在常规苏木精-伊红染色(HE)中出现伊红着色不均的原因分析。方法参照实用现代病理学技术的制片流程,对可能存在问题的组别一一排查。收集2023年8月至2024年1月中山市人民医院存档的120例胃肠活检石蜡标本切片,在染色不同时期进行上机HE染色,对比分析不同染色时期下的染色结果。结果排查后发现问题可能存在HE染色步骤组中,乙醇浓度下降对伊红着色有影响。设置3批片,在不同染色时期分别上机染色。镜下观察前、中、后3个染色时期中,伊红着色不均率分别为0%(0/40)、17.5%(7/40)、72.5%(29/40)。在不同染色时期中伊红着色不均率差异有统计学意义(χ^(2)=78.603,P<0.01)。结论在HE染色步骤中,机器在工作后期,随着染片工作量增大,脱水环节中乙醇浓度降低,对伊红着色有影响。Objective To explore the causes of uneven eosin staining in gastrointestinal biopsy tissues using hematoxylin eosin staining(HE).Methods According to the production process of practical modern pathological techniques,potential problematic groups were identified one by one.A total of 120 paraffin sections of gastrointestinal biopsy specimens archived by Zhongshan People′s Hospital were collected from August 2023 to January 2024,and machine HE staining was performed at different staining stages to compare and analyze the staining results at different staining stages.Results After investigation,it was found that the problem may be related to the HE staining step group,where the decrease in ethanol concentration has an impact on eosin staining.Three batches of slices were set up and stained by machine at different staining stages.During the three staining stages(before,during,and after),the uneven staining rates of eosin were 0%(0/40),17.5%(7/40),and 72.5%(29/40),respectively,through microscopic observation.There was a statistically significant difference in the uneven staining rate of eosin at different staining stages(χ^(2)=78.603,P<0.01).Conclusion In the HE dyeing process,during the later stage of the operation,as the workload of the dyeing slides increases,the ethanol concentration in the dehydration process decreases,which has an impact on the staining of eosin.
