摘  要:目的:通过建立大鼠幼鼠颅骨缺损模型(2mm、5mm),探讨硬脑膜在颅骨缺损中促进骨修复的作用,并观察不同大小颅骨缺损中硬脑膜促成骨基因表达差异,为在临床治疗小儿颅骨缺损中补充理论基础。方法:选取60只10日龄SD大鼠随机分为3组,在顶骨中心制造颅骨缺损,分别是2mm缺损组、5mm缺损组、未缺损对照组(其中2mm缺损组、5mm缺损组为实验组),每组20只。对照组暴露颅骨后不做处理。将每组大鼠随机分为两部分,分别在术后1周、术后6周取材。采用Western-blot、RT-qPCR检测不同时期颅骨缺损处硬脑膜中骨形态蛋白2(Bmp2)、Runt相关转录因子2(Runx2)、成纤维细胞生长因子受体2(FGFR2)的蛋白表达情况及mRNA表达情况。结果:(1)术后1周时,实验组Bmp2、Runx2、FGFR2表达量均高于未缺损对照组,其中2mm颅骨缺损组的Bmp2、Runx2蛋白表达量显著高于5mm缺损组,FGFR2蛋白表达量低于5mm缺损组(P<0.05)。术后6周时,实验组中Bmp2和Runx2蛋白表达量均高于未缺损对照组,其中5mm缺损组Bmp2、Runx2表达量高于2mm缺损组(P<0.05);术后6周时,2mm缺损组的FGFR2蛋白表达量与对照组无明显差异(P>0.05),但5mm缺损组的FGFR2蛋白表达量高于未缺损对照组和2mm缺损组(P<0.05)。(2)Runx2、Bmp2、FGFR2 mRNA表达水平情况:表达趋势同Western blot结果表达趋势相似。结论:(1)颅骨缺损后,未成熟硬脑膜促成骨因子表达增加;(2)硬脑膜中不同促成骨因子在不同大小的颅骨缺损中表达量存在差异。Objective:In order to supplement the theoretical basis for the clinical treatment of pediatric skull defects,the role of dura mater in promoting bone repair in skull defects was investigated by establishing skull defect models(2mm and 5 mm)in rat and young rat skull defects,and the differences in bone gene expression contributed by dura mater in different sizes of skull defects were observed.Methods:A total of 6010-day-old SD rats were randomly divided into three groups,and the skull defect was created in the center of the parietal bone,including 2mm defect group,5mm defect group,and non-defect control group,with 20 rats in each group(the 2mm defect group and the 5mm defect group were the experimental group).The non-defect control group was not treated after the skull was exposed.Rats in each group were randomly divided into two parts,which were collected at 1 week and 6 weeks after surgery.The protein expressions of bone morphin protein 2(Bmp2),Runt-related transcription factor 2(Runx2)and fibroblast growth factor receptor 2(FGFR2)in the dura mater of skull defects at different stages were analyzed by Western-blot and RT-qPCR.Results:(1)The expression levels of Bmp2,Runx2,and FGFR2 protein were higher in the experimental group than in the non-defect control group at 1 week after surgery(P<0.05).At 1 week after surgery,the expression levels of Bmp2 and Runx2 protein in the 2mm defect group were higher than those in the 5mm defect group,and the expression levels of FGFR2 protein were lower than those in the 5mm defect group(P<0.05).At 6 weeks after surgery,the expression of Bmp2 and Runx2 protein in the experimental group was higher than that in the non-defect control group,and the expression of Bmp2 and Runx2 protein in the 5mm defect group was higher than that in the 2mm defect group(P<0.05).At 6 weeks after surgery,there was no significant difference between in the 2mm defect group and non-defect control group of FGFR2 protein expression levels(P>0.05),while the expression levels of FGFR2 protein in the 5mm def
