四逆汤对A549肺癌干细胞增殖及凋亡的影响

摘　　要：目的研究经方四逆汤对A549肺癌干细胞增殖及凋亡的影响。方法利用无血清悬浮培养诱导A549肺癌干细胞,检测标志物CD133阳性率,验证细胞干性。分别用不同剂量四逆汤含药血清(10%、20%)干预A549肺癌干细胞。CCK-8、流式细胞检测四逆汤干预后对A549肺癌干细胞增殖及凋亡能力的影响。通过qPCR检测四逆汤干预后对A549肺癌干细胞增殖及凋亡能力的影响。结果经无血清培养基培养的细胞球CD133阳性率为(82.3667±0.47258)%,明显高于肺癌细胞组(P<0.05);经CCK-8检测四逆汤干预的肺癌干细胞组在48、72h时吸光值较对照组均显著降低(P<0.05),四逆汤干预的肺癌干细胞凋亡率较对照组显著升高(P<0.05);四逆汤含药血清可显著降低A549肺癌干细胞PCNA、Ki67和Bcl-2 mRNA水平(P<0.05),显著升高肺癌干细胞Bax mRNA水平(P<0.05)。结论四逆汤能抑制A549肺癌干细胞增殖并促进其凋亡。Objective:To study the effect of Sini Decoction on the proliferation and apoptosis of A549 lung cancer stem cells.Methods:A549 lung cancer stem cells were induced by serum-free suspension culture,and the positive rate of CD133 was detected to verify the cell stemness.The A549 lung cancer stem cells were intervened with different doses of herb-containing serum of Sini Decoction(10%,20%).The effect of Sini Decoction on the proliferation and apoptosis of A549 lung cancer stem cells was detected by CCK-8 and flow cytometry.And qPCR was used to detect the effect of Sini Decoction on the proliferation and apoptosis of A549 lung cancer stem cells.Results:The positive rate of CD133 was(82.3667±0.47258)%,which was significantly higher than that of lung cancer cell group(P<0.05).After CCK-8 detection,the light absorption value of lung cancer stem cells in the Sini Decoction intervention group was significantly decreased compared with that of the control group at 48 and 72h(P<0.05),and the apoptosis rate of lung cancer stem cells in the Sini Decoction intervention group was significantly increased compared with that of the control group(P<0.05).The herb-containing serum of Sini Decoction significantly decreased the mRNA levels of PCNA,Ki67 and Bcl-2 of A549 lung cancer stem cells(P<0.05),and significantly increased the mRNA levels of Bax of lung cancer stem cells(P<0.05).Conclusion:Sini Decoction can inhibit the proliferation and promote the apoptosis of A549 lung cancer stem cells.
