摘 要:目的探究木犀草素对正畸牙移动模型大鼠骨改建及骨形态发生蛋白2(BMP2)/Smad1信号通路的影响。方法建立大鼠正畸牙移动模型并分为模型组、木犀草素低(25 mg/kg)、中(50 mg/kg)、高(100 mg/kg)剂量组,每组12只;另取12只健康大鼠作为对照组。各组用相应药物进行灌胃,每天1次,连续30天。游标卡尺测量大鼠正畸牙移动距离;酶联免疫吸附法检测血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6水平;HE染色观察大鼠牙周组织改建情况;荧光定量PCR法检测大鼠牙周组织中BMP2、Smad1信使RNA(mRNA)水平;蛋白印迹法检测大鼠牙周组织中BMP2、Smad1、Runt相关转录因子2(RUNX2)、骨保护素(OPG)蛋白水平。结果对照组大鼠上颌骨结构正常。与对照组相比,模型组大鼠上颌骨压力侧牙周间隙变窄,上颌骨张力侧牙周韧带增宽,牙骨质表面可见较多的吸收陷窝,牙槽骨改建活跃,正畸牙移动距离、血清TNF-α、IL-1β、IL-6水平显著升高(P<0.05)。牙周组织BMP2、Smad1 mRNA和蛋白水平、OPG、RUNX2蛋白水平显著降低(P<0.05)。与模型组相比,木犀草素低、中、高剂量组大鼠上颌骨压力侧牙周间隙变窄逐渐明显,上颌骨张力侧新骨形成依次增多,骨吸收陷窝逐渐减少,血清TNF-α、IL-1β、IL-6水平依次降低(P<0.05)。正畸牙移动距离、牙周组织BMP2、Smad1 mRNA和蛋白水平、OPG、RUNX2蛋白水平依次升高(P<0.05)。结论木犀草素能加快正畸牙移动模型大鼠骨改建与骨形成过程,促进牙移动,抑制炎症反应,其机制可能与激活BMP2/Smad1信号通路有关。Objective To investigate the effects of luteolin on bone remodeling and bone morphogenetic protein 2(BMP2)/Smad1 signaling pathway in orthodontic tooth movement model rats.Methods The orthodontic tooth movement model was established and divided into model group,luteolin low(25 mg/kg),medium(50 mg/kg)and high(100 mg/kg)dose groups,with 12 rats in each group;another 12 healthy rats were taken as control group.Each group was given intragastric administration with corresponding drugs once a day for 30 days.The movement distance of orthodontic teeth was measured by vernier caliper;the serum levels of tumor necrosis factor(TNF)-α,interleukin(IL)-1βand IL-6 were detected by enzyme linked immunosorbent assay;HE staining was used to observe the periodontal tissue remodeling of rats;the mRNA levels of BMP2 and Smad1 in rat periodontal tissues were detected by fluorescence quantitative PCR;the protein levels of BMP2,Smad1,Runt-related transcription factor 2(RUNX2)and osteoprotegerin(OPG)in rat periodontal tissues were detected by Western blotting.Results The maxillary structure of rats in control group was normal.Compared with control group,the periodontal space on the pressure side of the maxilla in the model group was narrowed,and the periodontal ligament on the tension side of the maxilla was widened.More absorption lacunae were visible on the cementum surface,and alveolar bone remodeling was active.The orthodontic tooth movement distance,the serum levels of TNF-α,IL-1βand IL-6 were significantly increased(P<0.05).The mRNA and protein levels of BMP2 and Smad1,the protein levels of OPG and RUNX2 in periodontal tissues were significantly decreased(P<0.05).Compared with model group,the periodontal space narrowed gradually on the pressure side of the maxilla,the formation of new bone on the tension side increased successively,and the bone resorption lacunae gradually decreased in the luteolin low,medium and high dose groups.The levels of serum TNF-α,IL-1βand IL-6 were decreased in turn(P<0.05).The orthodontic tooth mov
