摘 要:本研究在单因素实验基础上采用正交试验优化,考察提取温度、提取时间和液料比对复方健脾养胃方中粗多糖提取率的影响,并考察纯化工艺对粗多糖保留率、浊度和感官评价的影响。结果表明,复方健脾养胃方中粗多糖的提取最优工艺为:95℃水浸提,液料比12∶1mL/g,提取时间2h,提取2次。优选的纯化工艺为:先将水提取液浓缩至相对密度为1.05~1.10(65℃),8000r/min离心20min后用孔径为0.45μm滤袋过滤,过滤液中粗多糖保留率为90.10%,浊度为31.4NTU,液体澄清透明,水提液稳定性好。On the basis of single factor experiment,orthogonal experimental optimization was used to investigate the effects of extraction temperature,extraction time,and liquid to material ratio on the extraction rate of crude polysaccharides in the compound Jianpi Yangwei fomula,and to investigate the effects of purification process on the retention rate,turbidity,and sensory evaluation of crude polysaccharides.The results showed that the optimal extraction conditions of crude polysaccharides were as follows:water extraction at 95℃,the ratio of liquid to material was 12∶1 mL/g,the extraction time was 2 h,and the extraction was twice.The optimum purification process was:the water extract was concentrated to a relative density of 1.05~1.10(65℃),centrifuged at 8000 r/min for 20 min,and then filtered with a filter bag with a pore size of 0.45μm.The retention rate of crude polysaccharide in the filtrate was 90.10%,the turbidity was 31.4 NTU,the liquid was clear and transparent,and the stability of the water extract was good.
