摘  要:【目的】基于外翻肠囊法研究乳脂肪主要成分的体外肠吸收特征。【方法】利用气相色谱-质谱联用仪(GC-MS)分析某乳品厂原料奶,获知其脂肪酸构成,再通过外翻肠囊实验法获取乳脂肪主要成分在大鼠不同肠段吸收转运数据,进而探讨不同肠段、不同给药时间和不同给药浓度对实验大鼠肠道乳脂肪主要成分吸收特性的影响。【结果】乳脂中共分析出26种脂肪酸,其中2,3,4-三甲基-戊酸的含量最高(54.68%),其次为4-甲基-庚酸(22.17%);2,3,4-三甲基-戊酸和4-甲基-庚酸的最佳给药部位分别为空肠和十二指肠,透过药量分别为31.08×10^(-3) mg、20.36×10^(-3) mg,单位时间内的吸收转化率分别为49.9993%、50.0043%,肠道剩余百分数分别为99.9931%、99.9913%。给药时间为120min时2,3,4-三甲基-戊酸的Ka、Papp分别为2.63、5.82×10^(-6),4-甲基-庚酸的Ka、Papp分别为4.65、1.26×10^(-6),均为最大值;在给药量为10.08mg/mL时,肠囊液中2,3,4-三甲基-戊酸的浓度为9.14×10^(-3)mg/mL,4-甲基-庚酸的浓度为6.36×10^(-3)mg/mL,均为最大值。【结论】2,3,4-三甲基-戊酸最佳吸收部位为空肠,4-甲基-庚酸最佳吸收部位为十二指肠;最佳时间为120 min,最佳浓度为10.08 mg/mL。【Objective】To study the in vitro intestinal absorption characteristics of the main components of milk fat based on the everted intestinal sac method.【Methods】Based on gas chromatography-mass spectrometry(GC-MS),the fatty acid composition of raw milk from a dairy plant was analysed,and then the absorption and transport data of the main components of milk fat in different intestinal segments of rats were obtained by the externally-turned-intestinal-bladder assay,so as to investigate the effect of different intestinal segments,different dosing times and different dosing concentrations on the absorption of the main components of milk fat from the intestinal tract of experimental rats.【Results】A total of 26 fatty acids were analyzed in milk fat,among which 2,3,4-trimethyl-pentanoic acid was the highest(54.68%),followed by 4-methyl-heptanoic acid(22.17%);the optimal administration sites for 2,3,4-trimethyl-pentanoic acid and 4-methyl-heptanoic acid were the jejunum and the duodenum,with the transmitting doses of 31.08×10^(-3) mg and 20.36×10^(-3) mg,the absorption conversion rate per unit time was 49.9965%,50.0043%,and the intestinal residual percentage was 99.9931%and 99.9913%,respectively;When the administration time was 120 minutes,the Ka and Papp of 2,3,4-trimethyl-pentanoic acid were 2.63 and 5.82×10^(-6),respectively,and the Ka and Papp of 4-methyl-heptanoic acid were 4.65and 1.26×10^(-6),respectively,all of which were the highest;The concentrations of 2,3,4-trimethyl-pentanoic acid in the intestinal vesicle fluid were 9.14×10^(-3) mg/mL,and 4-methyl-heptanoic acid was 6.36×10^(-3) mg/mL at a dosage of 10.08 mg/mL,both of them were maximum.【Conclusion】The optimal absorption site for 2,3,4-trimethyl-pentanoic acid was the jejunum,and the optimal absorption site for 4-methyl-heptanoic acid was the duodenum;the optimal time was 120 minutes,and the optimal concentration was 10.08 mg/mL.
