摘  要:目的:探讨赖氨酸特异性去甲基化酶(lysine [K]-specific demethylase 6B,KDM6B)和叉头框蛋白O1(forkhead box protein O1,FOXO1)在浆液性卵巢癌(serous ovarian cancer,SOC)中的表达水平及其与患者临床病理特征和预后的关系。方法:纳入我院妇科2018年04月至2023年04月收治入院接受手术治疗并经过病理证实的367例SOC患者和150例卵巢良性肿瘤患者。采用免疫组织化学法检测KDM6B、FOXO1蛋白在SOC组织中的表达水平,并分析二者表达水平与临床病理特征的关系。应用Pearson相关系数分析SOC组织中KDM6B与FOXO1蛋白表达的相关性。利用Kaplan-Meier法和Log-rank检验对不同KDM6B、FOXO1蛋白表达水平的SOC患者预后的差异进行比较。结果:KDM6B与FOXO1蛋白SOC组织中高表达率分别为48.77%和53.13%,显著高于正常卵巢组织的21.33%和32.00%(P均<0.05)。不同KDM6B蛋白表达水平的SOC患者在年龄、肿瘤分化程度、有无淋巴结转移、FIGO分期的差异具有统计学意义(P<0.05),FOXO1蛋白表达水平与患者肿瘤分化程度、有无淋巴结转移、FIGO分期有关(P<0.05)。SOC组织KDM6B与FOXO1蛋白表达呈正相关(r=0.668,P<0.05)。KDM6B、FOXO1蛋白高表达者的总生存率和无进展生存率均显著低于低表达者(P<0.05)。结论:KDM6B、FOXO1蛋白在SOC组织中高表达,二者表达水平呈正相关,且KDM6B、FOXO1蛋白高表达者预后较低表达者更差,提示二者均可能在SOC发生发展中发挥促癌作用,且可作为影响SOC患者预后的关键因素,可能是潜在的SOC早期诊断和预后评估标志物。Objective:To investigate the expression of lysine [K]-specific demethylase 6B(KDM6B) and forkhead box protein O1(FOXO1) in serous ovarian cancer(SOC) tissues,and to analyze their relationships with clinical pathological characteristics and prognosis of SOC patients.Methods:A total of 367 SOC patients and 150 patients with ovarian benign tumor admitted to our hospital from april 2018 to april 2023 for surgical treatment and confirmed by pathology were included.Immunohistochemical methods were conducted to detect the expressions of KDM6B and FOXO1 proteins in SOC tissues,and the correlations between their expressions and clinical pathological characteristics were analyzed.Pearson correlation coefficient was used to analyze the correlation between KDM6B and FOXO1 protein expression in SOC tissues.The prognosis of SOC patients with different expressions of KDM6B and FOXO1 protein was compared using Kaplan-Meier method and Log-rank test.Results:The high expression rates of KDM6B and FOXO1 protein in SOC tissue were 48.77% and 53.13%,which were significantly higher than those in normal ovarian tissues(21.33% and 32.00%,P<0.05),respectively.There were significant differences in age,tumor differentiation,lymph node metastasis,and FIGO stage in SOC patients with different expressions of KDM6B protein(P<0.05),while the expressions of FOXO1 protein was correlated with tumor differentiation,lymph node metastasis,and FIGO stage(P<0.05).There was a positive correlation between KDM6B and FOXO1 protein expression in SOC tissues(r=0.668,P<0.05).The overall survival rate and progression free survival rate of individuals with high expression of KDM6B and FOXO1 proteins were significantly lower than those with low expression(P<0.05).Conclusion:KDM6B and FOXO1 proteins are highly expressed in SOC tissues,and their expressions are positively correlated.Moreover,patients with high expression of KDM6B and FOXO1 proteins have a poorer prognosis compared to those with low expression,suggesting that the KDM6B and FOXO1 may play roles in pr
