摘　　要：目的:检测着丝粒蛋白A(centromeric protein A,CENP-A)在膀胱癌组织中的mRNA和蛋白表达水平,分析其临床意义及与患者临床病理特征的相关性。方法:RT-qPCR技术检测30例膀胱癌患者外周血单个核细胞中CENP-A mRNA的表达水平;免疫组化方法检测45例膀胱癌组织中CENP-A蛋白的表达水平及其与临床病理特征的关系;ROC曲线分析CENP-A蛋白表达情况对于膀胱癌的诊断价值。结果:与对照组相比,膀胱癌组织中CENP-A的mRNA及蛋白表达显著上调,CENP-A的表达量与年龄、性别等无相关性,与肿瘤分期及肿瘤恶性程度呈正相关。对其进行ROC曲线分析,发现CENP-A对膀胱癌诊断的敏感度和特异度较高,具有较好的临床价值。结论:CENP-A在膀胱癌组织中表达上调且与膀胱癌患者的分期情况相关,可能成为膀胱癌精准分子诊断的潜在靶基因,在其诊断治疗中发挥重要的作用。Objective:To detect the mRNA and protein expression levels of centromeric protein A(CENP-A) in bladder cancer tissues,and analyze its clinical significance and correlation with relevant pathological characteristics.Methods:The expression of CENP-A mRNA in peripheral blood mononuclear cells of 30 patients with bladder cancer was detected by RT-qPCR.The expression of CENP-A protein in 45 cases of bladder cancer and its relationship with clinicopathological characteristics were detected by immunohistochemistry.ROC curve analysis of CENP-A protein expression for the diagnosis of bladder cancer.Results:Compared with the control group,the mRNA and protein expression of CENP-A in bladder cancer tissues was significantly up-regulated.The expression of CENP-A was not correlated with age,gender,etc.,but was positively correlated with tumor stage and tumor grade.The ROC curve analysis showed that CENP-A had high sensitivity and specificity in the diagnosis of bladder cancer,and had good clinical value.Conclusion:The expression of CENP-A is up-regulated in bladder cancer tissues and is related to the stage of bladder cancer patients,which may become a potential target gene for accurate molecular diagnosis of bladder cancer and play an important role in its diagnosis and treatment.
