摘 要:目的:探讨有氧匀速运动和高强度间歇性运动对雄性小鼠抑郁样行为和脑内神经肽相关基因表达的影响。方法:将18只小鼠随机分为对照组、匀速运动组和高强度间歇性运动组,每组6只,除对照组外其余小鼠连续运动8周。运动干预后,采用强迫游泳实验检测小鼠不动时间,实时荧光定量PCR检测小鼠脑组织不同部位(前额皮层、纹状体、海马、下丘脑)神经肽相关基因的表达,即脑源性神经营养因子(Bdnf)、饥饿激素(Ghrelin)、神经肽Y(Npy)、刺鼠关联蛋白(Agrp)、可卡因-苯丙胺调节转录肽(Cart)、前阿黑皮素原(Pomc)、促肾上腺皮质激素释放激素(Crh)、爱帕琳肽(Apelin)和黑色素聚集激素(Mch)。结果:与对照组相比,两种运动组小鼠的不动时间均显著降低(P<0.05),高强度间歇性运动组小鼠不动时间降低更多;匀速运动组小鼠下丘脑Apelin基因的表达水平显著升高(P<0.01);高强度间歇性运动组小鼠纹状体Apelin基因的表达水平显著降低(P<0.05),下丘脑Crh基因(P<0.05)和Cart基因的表达水平(P<0.01)均显著降低。结论:有氧匀速运动和高强度间歇性运动均在一定程度上改善小鼠抑郁样行为,后者效果更好。两种运动方式均可影响小鼠纹状体和下丘脑Apelin基因表达,高强度间歇性运动还影响小鼠下丘脑Crh和Cart基因表达。Objective:To investigate the effects of aerobie exercise training and high intensity interval training on depression-like behavior and neuropeptide related genes expression in male mice.Methods:18 mice were randomly divided into control group,constant speed exercise group and high intensity intermittent exercise group,with 6 mice in each group.Except for mice in the control group,the remaining mice exercised continuously for 8 weeks.After exercise intervention,the immobility time of mice was detected by forced swimming test,and the expression of neuropeptide-related genes in different tissues of the brain(prefrontal cortex,striatum,hippocampus,hypothalamus)were detected by reverse transcription and fluorescence quantitative PCR,including Bdnf,Ghrelin,Npy,Agrp,Cart,Pomc,Crh,Apelin and Mch.Results:Compared with the control group,the immobility time of mice in both exercise groups was decreased(P<0.05),and the immobility time of mice in high-intensity intermittent exercise group decreased stronger.The expression of A pelin mRNA in hypothalamus of mice in constant speed exercise group was significantly increased(P<0.01).For high intensity intermittent exercise group,the A pelin mRNA was significantly decreased(P<0.05)in striatum,and the mRNA level ofCrh(P<0.05)andCart(P<0.01)were significantly decreased in hypothalamus,compared with control group.Conclusion:Both constant speed exercise training and high intensity interval training could alleviate depression-like behavior in mice,and the high-intensity interval training is better.Both types of exercise influenced the mRNA level of Apelin in the striatum and hypothalamus in mice,and high intensity interval training reduced the Crh and Cart genes expression in the hypothalamus.
