摘 要:目的探究重组香脂素对宫颈癌细胞的细胞周期、Bax、caspase-3、Bcl-2调控及作用机制。方法将4株宫颈癌海拉细胞株培养后分为44个单独样本,其中11株作为对照,进行常规培养,其余33株为研究组,分别给予10、50、100μg/mL重组香脂素进行干预,观察各组样本细胞凋亡、细胞周期、caspase活性、Bax、caspase-3、Bcl-2基因表达及GPR78、CHOP基因表达。结果研究组重组香脂素干预后海拉细胞细胞存活率显著低于对照组,且3组间比较差异具有统计学意义(P<0.05)。DAPI染色后显示,重组香脂素处理的细胞显示出凋亡小体(细胞核破碎和凝聚),而未处理或对照细胞未显示出明显的细胞凋亡;研究组G0/G1周期细胞占比显著高于对照组,S、M周期细胞占比显著低于对照组且呈现剂量依赖性(P<0.05);研究组caspase-3、caspase-8、caspase-9活性和Bax、casepase-3、GPR78、CHOP基因相对表达量显著高于对照组,Bcl-2基因相对表达显著低于对照组,且呈现剂量依赖性变化(P<0.05)。结论重组香脂素能通过调控宫颈癌细胞细胞周期及caspase活性、Bax、caspase-3、Bcl-2基因表达促进宫颈癌细胞凋亡,同时其还能调控GPR78、CHOP基因表达及诱导内质网应激激活影响患者病程。Objective To explore the regulation and mechanism of recombinant balsam on cell cycle,Bax,caspase-3 and Bcl-2 of cervical cancer cells.Methods 4 cervical cancer Hela cell lines were cultured and divided into 44 individual samples,11 of which were used as control for routine culture,and the remaining 33 were used as study group for intervention with 10,50,100μg/mL recombinant balsamin,respectively.Apoptosis,cell cycle,caspase activity,gene expression of Bax,caspase-3,Bcl-2 and gene expression of GPR78 and CHOP were observed.Results The survival rate of Hela cells in the study group was significantly lower than that in the control group,and the difference among the three groups was statistically significant(P<0.05).After DAPI staining,cells treated with recombinant balsamin showed apoptotic bodies(nucleus fragmentation and aggregation),while untreated or control cells did not show obvious apoptosis.The proportion of G0/G1 cycle cells in the study group was significantly higher than that in the control group,and the proportion of S and M cycle cells was significantly lower than that in the control group in a dose-dependent manner(P<0.05).The activities of caspase-3,caspase-8 and caspase-9 and the relative expression levels of Bax,casepase-3,GPR78 and CHOP genes in the study group were significantly higher than those in the control group,while the relative expression levels of Bcl-2 genes in the study group were significantly lower than those in the control group,with dose-dependent changes(P<0.05).Conclusion Recombinant balanin can promote apoptosis of cervical cancer cells by regulating cell cycle,caspase activity,Bax,caspase-3 and Bcl-2 gene expression,and can also regulate the expression of GPR78 and CHOP genes and induce endoplasmic reticulum stress activation.
