摘 要:目的:优化天仙果总黄酮提取工艺,并测定其抗氧化活性。方法:通过响应面优化建立数学模型,考察乙醇体积分数、料液比、提取时间、提取次数4个因素,得到天仙果总黄酮最优提取条件。考察天仙果抗氧化活性,进行1,1-二苯基-2-三硝基苯肼(DPPH)法、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)法、羟自由基清除能力的测定。结果:天仙果总黄酮最佳提取工艺为乙醇体积分数60%、料液比1∶23、提取时间18 min、提取1次,在此条件下,提取率为16.25 mg·g^(-1),天仙果总黄酮对DPPH、ABTS、羟自由基清除率分别为92.84%、89.34%、69.73%。结论:优化后的提取工艺可操作性强、稳定可靠,天仙果抗氧化活性好。Objective:To optimize the extraction process and determine the antioxidant activity of total flavonoids from Ficus erecta(Thunb.)var.beecheyana(Hook.et Arn.)King.Methods:Four factors,namely ethanol concentration,material-liquid ratio,extraction time,and number of extractions,were selected to obtain the optimal conditions through response surface methodology and mathematical modeling,thus improving the extraction rate of total flavonoids from F.erecta.The in vitro antioxidant activity was investigated,measuring 1,1-Diphenyl-2-picrylhydrazyl radical(DPPH),2,2-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS),and hydroxyl radical scavenging capacities.Results:The optimal extraction process for total flavonoids from F.erecta was identified with 60%ethanol,1:23 material-liquid ratio,and 18-min extraction time,under which the extraction rate was 16.25 mg·g^(-1).The scavenging rates of total flavonoids from F.erecta for DPPH,ABTS,and hydroxyl radicals reached 92.84%,89.34%,and 69.73%,respectively.Conclusion:The optimized extraction process was operable,stable,and reliable,exhibiting good antioxidant activity.
