摘 要:目的探讨脂肪间充质干细胞(ADSC)脑内定向移植治疗大鼠脑出血的机制。方法对从大鼠腹股沟处吸脂来源的脂肪组织进行ADSC的分离、传代培养、鉴定、冻存和复苏、体外诱导分化及储存。建立大鼠脑出血微创抽吸术模型30只,采用抽签法将其分为三组,A组:磷酸盐缓冲液3 ml;B组:ADSC 1×10^(8)悬浮于3 ml磷酸盐缓冲液;C组:ADSC诱导分化的神经干细胞1×10^(8)悬浮于3 ml磷酸盐缓冲液。采用ADSC脑内定向治疗脑出血,通过Rogers Scale积分进行神经行为功能评价,在第1、3、5、7周进行,进行统计分析。通过超速离心法从血清中提取外泌体,分析其在三组大鼠中的差异。结果三组大鼠均出现不同程度的神经功能障碍,在第1、3、5、7周C组大鼠Rogers Scale评分均低于同时间的A、B组,差异有统计学意义(P<0.05);C组大鼠同组比较第5、7周Rogers Scale评分低于第1周,差异有统计学意义(P<0.05);在第1、3、5、7周A、B两组大鼠血清外泌体浓度均低于同时间的C组,差异有统计学意义(P<0.05)。结论ADSC通过脑内定向移植能够减轻脑出血大鼠模型症状,可能与其分泌外泌体有关。Objective To discuss adipose-derived stem cells(ADSC)through brain targeted transplantation treating rats with intracerebral hemorrhage and its mechanism.Methods Fatty tissue for liposuction sources from rats was got and ADSC were separated.ADSC were subcultured appraised cryogenic storaged recovered induced differentiation in vitro and stored.Thirty models of mice cerebral hemorrhage minimally invasive pumping were made,they were divided into three groups from lottery method,group A:3 ml phosphate buffer;group B:1×10^(8) cells of ADSC suspended in 3 ml phosphate buffer;group C:1×10^(8) cells of nerve stem induced and differentiated from ADSC suspended in 3 ml phosphate buffer.Rats of cerebral hemorrhage were treated using brain targeted therapy,statistical analysis was performed through the Rogers Scale points,and statistical analysis was performed on the 1st,3rd,5th weeks.By super-speed centrifugal method extracted exosomes from the serum,the differences between its three groups of rats were analyzed.Results All three groups of rats had different degrees of neurological dysfunction.In the 1st,3rd,5th,and 7th weeks,the Rogers Scale score was lower than that of group A and B,the difference was statistically significant(P<0.05).The Rogers Scale score of the 5th and 7th weeks of the rats group C was less than the 1st week,and the differences was statistically significant(P<0.05).In the 1st,3rd,5th,and 7th weeks,the concentration of A and B groups rats exosomes was lower than group C at the same time,and the difference was statistically significant(P<0.05).Conclusion ADSC can reduce the symptoms of cerebral hemorrhagic rats model through the directional transplantation of the brain,which may be related to the secretion of exosomes.
