摘 要:目的研究不同褐藻来源β-葡聚糖的结构及免疫活性差异,为β-葡聚糖药物的开发提供合适的藻源。方法通过热水提取、乙醇沉淀和柱层析分离从6种褐藻中制备β-葡聚糖,并利用高效液相色谱(HPLC)、红外光谱(IR)、核磁共振波谱(^(1)H NMR,^(13)C NMR和^(1)H^(13)C HSQC)对其结构进行表征。通过小鼠巨噬细胞RAW264.7吞噬中性红及NO释放实验评价其免疫调节活性。结果本文制备了6种结构不同的β-葡聚糖,对其结构分析,证明其含有β-1,3和β-1,6连接,相对分子质量为2 290~5 490,海茸中β-葡聚糖的含量最高且具有最优的免疫活性,是β-葡聚糖药物开发的良好资源。Objective To investigate the structure and immunological activity ofβ-glucan from six kinds of brown algae,and provide the algae source for the development of marine carbohydrate-based drugs.Methodsβ-glucans were prepared from 6 kinds of brown algae by hot water extraction,ethanol precipitation and column chromatography.Their structures were characterized by high performance liquid chromatography(HPLC),infrared radiation(IR)and nuclear magnetic resonance spectroscopy(^(1)H NMR,^(13)C NMR and ^(1)H^(13)C HSQC),and immune activities were evaluated by phagocytosis of neutral red and NO release amount of mouse macrophage RAW264.7.Results In this paper,six kinds ofβ-glucans with different structures were prepared,and their structures were analyzed.It was proved that they containedβ-1,3 andβ-1,6 linkages,and their relative molecular weights were 2290~5490,Durvillaea antarctica,which has the highest content ofβ-glucan and the best immune activity,is an ideal resource forβ-glucan drug development.
