摘　　要：目前慢性肝病已成为全球范围内高发病率和高病死率的疾病之一。维生素A(Vit A)是人体必需的脂溶性维生素之一,其活性代谢产物视黄酸与视黄酸受体结合,通过视黄酸信号通路发挥重要的生理作用。肝脏是Vit A储存和代谢的主要器官。肝脏Vit A含量减少、代谢异常和视黄酸信号转导异常与多种肝脏疾病有关,而恢复Vit A相关信号转导、使用天然或合成类视黄醇以及针对特定视黄酸受体的新型激动剂显示出治疗前景,但仍有许多机制尚不完全清楚。本文围绕Vit A在慢性肝病发病机制和治疗中的作用进行综述,以期为慢性肝病的基础研究和临床治疗提供新的视野。Chronic liver diseases(CLDs)have become one of the diseases with high morbidity and mortality worldwide.Vitamin A(Vit A)is an essential fat-soluble vitamin.Retinoic acid(RA),an active metabolite of Vit A,is a potent regulator of numerous biological pathways via the RA signaling pathway acting as binding to RA receptors(RARs).Liver is the primary organ for Vit A storage and metabolism in humans.A body of evidence shows that reductions in liver Vit A,aberrant Vit A metabolism,and dysregulated in RA signaling are implicated in numerous diseases of liver.Conversely,restoration of RA signaling,pharmacological treatments with natural and synthetic retinoids,and newer agonists for specific RARs show promising benefits for treatment of a number of liver diseases.However,the pathogenesis remains incompletely understood.Here we provide a comprehensive review of the literature demonstrating a role for Vit A in limiting the pathogenesis of these diseases and in the treatment of liver diseases,in order to provide a new perspective for research and treatment of CLDs.
