摘  要:目的优化食管鳞癌类器官的培养方法。方法收集郑州大学第二附属医院胸外科2023年5月至2024年3月食管鳞癌手术标本20例,用含/不含R-spondin 1的培养基培养类器官,观察二者对类器官生长的影响,同时比较市售的0.25%Trypsin-EDTA、1×Tryple和自配的组织消化液对类器官的消化效果。结果用含/不含R-spondin 1培养基培养食管鳞癌类器官时,两者的类器官大小、生长速率无明显差别(P>0.05)。0.25%Trypsin-EDTA消化细胞球时,细胞分散状态好,所生成的细胞球的数量均多于组织消化液和1×Tryple,1×Tryple消化细胞球的效率和所生成的细胞球的数量均大于组织消化液。结论R-spondin 1对食管鳞癌类器官的形成无明显影响;类器官传代扩增时,用0.25%Trypsin-EDTA有较好的消化效果。Objective To optimise the culture method of oesophageal squamous carcinoma organoids.Methods Twenty surgical specimens of oesophageal squamous carcinoma were collected from the Department of Thoracic Surgery of the Second Affiliated Hospital of Zhengzhou University from May 2023 to Mar.2024,and the effects on the growth of organoids were observed with medium with/without R-spondin 1.Meanwhile,the digestion effects of the commercially available 0.25%Trypsin-EDTA,1×Tryple and the self-prepared tissue digestive solution on the organoids were compared.Results No significant difference in the size and growth rate of oesophageal squamous carcinoma organoids was observed when cultured with or without R-spondin 1(P>0.05).When 0.25%Trypsin-EDTA effectively digested cell spheroids into a well-dispersed state,a greater number of cell spheroids generated compared to tissue digestion solution and 1×Tryple.The efficiency of cell spheroids digestion and the number of cell spheroids generated were higher with 1×Tryple compared to tissue digestion solution.Conclusion R-spondin 1 did not significantly affect organoid formation.For passaging and expansion of organoids,0.25%Trypsin-EDTA exhibited superior digestion efficacy.
