摘 要:目的分析乳腺癌组织细胞周期蛋白B1(CCNB1)、同源盒蛋白A7(HOXA7)表达水平及临床意义。方法收集2019.01—2020.10南通市中医院收治的53例乳腺癌患者术中切除的癌组织及癌旁组织,采用qRT-PCR检测CCNB1、HOXA7 mRNA表达水平,采用免疫组化(IHC)检测CCNB1、HOXA7蛋白表达水平,收集患者临床病理特征,采用Kaplan-Meier法分析CCNB1、HOXA7表达水平与患者3年生存率的相关性,采用多因素Cox分析影响患者预后的因素。结果在癌组织中,CCNB1、HOXA7的阳性表达率分别为64.15%、66.04%,CCNB1、HOXA7的阳性表达率、mRNA表达水平显著高于癌旁组织(P<0.05);CCNB1、HOXA7表达与肿瘤直径、TNM分期、雌激素受体状态有关(P<0.05);且肿瘤直径≥2 cm、TNM分期Ⅲ+Ⅳ期、雌激素受体状态阳性以及CCNB1、HOXA7阳性表达均为影响患者预后的危险因素(P<0.05);3年内,CCNB1阳性表达患者生存率(61.76%)低于阴性表达患者(89.47%)(P<0.05);HOXA7阳性表达患者生存率(60.00%)低于阴性表达患者(94.44%)(P<0.05)。结论在乳腺癌组织中,CCNB1、HOXA7表达显著升高,是乳腺癌患者预后的影响因素。Objective To study the expression levels of cyclin B1(CCNB1)and homeobox A7(HOXA7)in breast cancer tissues and their clinical significance.Methods The cancerous and paracancerous tissues of 53 patients with breast cancer who were treated in Nantong Hospital of Traditional Chinese Medicine from September 2019 to October 2020 were collected,qRT-PCR was applied to detect the expression levels of CCNB1 and HOXA7 mRNA,IHC was applied to detect the expression levels of CCNB1 and HOXA7 proteins,the general clinicopathological characteristics of breast cancer patients were collected,Kaplan-Meier method was applied to analyze the relationship between the expression levels of CCNB1 and HOXA7 and the three-year survival status of patients,multivariate Cox was used to analyze the factors affecting the prognosis of breast cancer patients.Results In cancer tissues,the positive expression rates of CCNB1 and HOXA7 were 64.15%and 66.04%,respectively,and the positive expression rates and mRNA expression levels of CCNB1 and HOXA7 were significantly higher than those of paracancerous tissues(P<0.05).In breast cancer patients,the expression of CCNB1 and HOXA7was related to tumor diameter,TNM stage and estrogen receptor status(P<0.05).And tumor diameter≥2 cm,TNM stageⅢ+Ⅳ,positive estrogen receptor status,and positive expression of CCNB1 and HOXA7 were all risk factors affecting patient prognosis(P<0.05).The three-year survival rate of patients with positive expression of CCNB1(61.76%)was lower than that of patients with negative expression of CCNB1(89.47%)(P<0.05).The three-year survival rate(60.00%)of HOXA7 positive expression patients was lower than that of HOXA7 negative expression patients(94.44%)(P<0.05).Conclusion In breast cancer tissue,the expression of CCNB1 and HOXA7 is obviously increased,and are influential factors for prognosis of breast cancer patients.
