菊粉重塑肠道菌群改善2型糖尿病db/db小鼠作用机制的研究

摘  要:目的分析菊粉通过重塑肠道菌群改善2型糖尿病(T2DM)的作用。方法选用20只db/db瘦素受体突变小鼠作为T2DM模型,小鼠空腹血糖≥11.1 mmol/L为糖尿病,再随机分为无干预糖尿病组(DM组,n=10)和菊粉干预糖尿病组(INU/DM组,n=10)。10只C57BL/6J小鼠为阴性对照组。INU/DM组小鼠每日每只以5%(wt/wt)菊粉连续灌胃6周,对照组和DM组小鼠每日给予等量的生理盐水灌胃6周。分别检测各组小鼠体重、血糖、糖化血红蛋白、血脂及脂多糖(LPS)水平。采用16S rRNA测序分析各组肠道菌群变化。结果INU/DM组与DM组相比,体重增加平稳,血糖、甘油三酯及总胆固醇明显下降(P<0.05)。INU/DM组小鼠血浆LPS较DM组明显降低(P<0.05)。16S rRNA测序分析,发现补充菊粉能够显著改变T2DM小鼠肠道菌群多样性和组成,增加了蓝藻菌门和产短链脂肪酸(SCFAs)拟杆菌科及属水平的相对丰度(P<0.05),同时降低了致病菌脱铁杆菌门、软壁菌门、瘤壁球菌科及反刍螺菌_6属的相对丰度(P<0.05)。结论菊粉通过重塑肠道菌群并抑制LPS易位控制T2DM。Objective To investigate the effect of inulin on T2DM via refactoring gut microbiota in mice.Methods 20 db/db leptin receptor mutant mice were used as the T2DM,According to previous research methods,model mice were randomly allocated into inulin treated group(INU/DM group,n=10)and non-treated group(DM group,n=10)and 10 C57BL/6J mice were used as the negative control group.5%(wt/wt)inulin was orally administered to the treated groups for six weeks,and an equal volume of saline was orally administered to the untreated groups.Body weight,blood glucose,glycated hemoglobin,blood lipids and LPS levels were detected in each group.16S rRNA sequencing was used to analyze the changes of gut microbiota.Results Blood glucose,glycated hemoglobin,blood lipids and LPS levels were significantly decreased in INU/DM group compared to DM group(P<0.05).Declined LPS level was observed in INU/DM group compared with that in DM group(P<0.05).The supplementation of inulin significantly altered the beta diversity and relative abundance of the gut microbiota by increasing the relative abundance of anti-diabetic cyanobacteria and SCFAs-producing bacteroidaceae and genus levels(P<0.05).Concurrently,inulin reduced the relative abundances of pathogenic bacteria including deferribacteres,tenericutes,ruminococcaceae,and ruminclostridium_6(P<0.05).Conclusion Inulin could ameliorates T2DM by reshaping the gut microbiota and inhibiting LPS translocation.
