摘  要:目的:基于生物信息学探讨蛋白脂质蛋白2(PLP2)在肝癌(LIHC)中的表达,并用于评估LIHC患者预后的价值及与免疫浸润的相关性。方法:通过GEPIA和UALCAN数据库分析PLP2在LIHC中的表达、与肿瘤分级分期相关性以及与肿瘤患者预后的相关性。通过TIMER数据库分析LIHC中PLP2表达水平与免疫细胞浸润水平的关系。通过String数据库分析与PLP2相关蛋白的互作网络。结果:PLP2在LIHC中的表达明显高于正常组织;PLP2与肿瘤病理分级以及临床分期具有一定的相关性;PLP2的高表达水平与LIHC患者的预后呈负相关,PLP2表达越高,预后越差;PLP2的表达水平与B细胞、CD4^(+)T细胞、巨噬细胞、CD8^(+)T细胞、树突状细胞、中性粒细胞的免疫浸润水平呈正相关。PLP2可能通过与TCP1、PLP1、IFR3、BECN1、PDCL2等蛋白的相互作用调控了LIHC的发生发展。结论:在LIHC患者中,PLP2的高表达与预后以及免疫浸润正相关,是LIHC患者预后的危险因素,有望成为LIHC免疫细胞浸润和预后相关的生物学标志物。Objective:To explore the expression of proteolipid protein 2(PLP2)in Liver hepatocellular carcinoma(LIHC)based on bioinformatics,and to evaluate the prognostic value of LIHC patients and their correlation with immune infiltration.Methods:The expression of PLP2 in LIHC,its correlation with tumor grading and staging,and its correlation with tumor patient prognosis were analyzed using GEPIA and UALCAN databases.Analyze the relationship between PLP2 expression level and immune cell infiltration level in LIHC through TIMER database.Analyze the interaction network with PLP2 related proteins through the String database.Results:The expression of PLP2 in LIHC was significantly higher than that in normal tissues.PLP2 has a certain correlation with tumor pathological grading and clinical staging.The high expression level of PLP2 is negatively correlated with the prognosis of LIHC patients.The higher the expression of PLP2,the worse the prognosis.The expression level of PLP2 is positively correlated with the immune infiltration level of B cells,CD4^(+)T cells,macrophages,CD8^(+)T cells,dendritic cells,and neutrophils.PLP2 may regulate the occurrence and development of LIHC through interactions with proteins such as TCP1,PLP1,IFR3,BECN1,PDCL2,etc.Conclusion:In LIHC patients,high expression of PLP2 is positively correlated with prognosis and immune infiltration,and is a risk factor for the prognosis of LIHC patients.It is expected to become a biological marker for immune cell infiltration and prognosis in LIHC.
