摘 要:目的此研究旨在通过网络药理学探讨丹参在治疗血管神经性头痛中的有效成分、作用靶点以及相关途径。方法对丹参主要成分和作用靶点进行检索,采用中药系统药理学数据库与SwissTargetPrediction网站筛选出丹参的活性成分及靶点,在人类基因数据库GeneCards中检索血管神经性头痛相关靶点,取药物与疾病的共同靶点绘制韦恩图,通过STRING在线分析平台分析药物与疾病共同靶点,进行蛋白质-蛋白质相互作用网络构建,利用Metascape数据库和微生信平台对药物与疾病共同靶点进行GO功能富集分析及KEGG通路富集分析。结果丹参筛选出21个有效化学成分及397个作用靶点,Venny图结合Cytoscape分析结果显示54个靶点与血管神经性头痛疾病靶点有交集,其中有11个核心靶点;GO分析和KEGG结果显示,核心靶点参与了多条功能富集及多条通路富集,其中主要为细胞膜及钙离子通路相关的富集结果。结论丹参对血管神经性头痛可能通过多成分、多靶点、多通路参与其治疗,主要机制可能为参与钙离子通路转运等。Objective The purpose of this study was to explore the effective components,targets and related pathways of Danshen in the treatment of angioneurotic headache through network pharmacology.Methods The main components and targets of S.miltiorrhiza were searched.The active components and targets of S.miltiorrhiza were screened out by using the Traditional Chinese Medicine system pharmacology database and SwissTargetPrediction website.The targets related to angioneurotic headache were searched in the human gene database GeneCards.The common targets of drugs and diseases were used to draw the Venn diagram.The common targets of drugs and diseases were analyzed by STRING online analysis platform,and the protein-protein interaction network was constructed.The GO function enrichment analysis and KEGG pathway enrichment analysis of the common targets of drugs and diseases were carried out by using Metascape database and micro-bioinformatics platform.Results 21 effective chemical components and 397 targets were screened out from Salvia miltiorrhiza.Venny diagram combined with Cytoscape analysis showed that 54 targets intersected with the targets of angioneurotic headache disease,including 11 core targets.GO analysis and KEGG results showed that the core targets were involved in multiple functional enrichment and multiple pathway enrichment,mainly related to cell membrane and calcium ion pathway.Conclusion Salvia miltiorrhiza may participate in the treatment of angioneurotic headache through multi-component,multi-target and multi-pathway.The main mechanism may be to participate in the transport of calcium ion pathway.
