摘　　要：小泛素样修饰(SUMO)是一种动态可逆的翻译后修饰,在体内多个水平上调节着蛋白质功能。研究结果显示一些关键肿瘤因子和肿瘤抑制因子是SUMO底物。在肿瘤的环境中,SUMO通路成员不仅生物活性由于条件的改变(如缺氧)而发生改变外,而且在不同肿瘤中的表达水平也会出现异常。SUMO通路表达失调必将破坏SUMO化与去SUMO化之间的动态平衡,该平衡一旦被打破,将促进各种肿瘤的发生和耐药。现已研究表明许多与特定肿瘤发病机制相关的分子机制都涉及SUMO化修饰,这也突出了SUMO通路作为恶性肿瘤治疗靶点的潜在可能性。本文总结SUMO化修饰通路在常见恶性肿瘤中的靶向治疗及其作用机制的最新研究进展。Small ubiquitin-like modifier(SUMO)is a dynamically reversible post-translational modification that regulates protein function at multiple levels in the body.Research has found that some key tumor factors and tumor suppressors are SUMO substrates.In the tumor environment,the SUMO pathway members not only exhibit changes in biological activity due to changes in conditions such as hypoxia,but also exhibit abnormal expression levels in different tumors.The imbalance of SUMO pathway expression will inevitably disrupt the dynamic balance between SUMOylation and de-SUMOylation.Once this balance is broken,it will promote the occurrence and drug resistance of various tumors.Research has shown that many molecular mechanisms related to specific tumor pathogenesis involve SUMO modification,highlighting the potential of the SUMO pathway as a therapeutic target for malignant tumors.Here,we comprehensively and systematically summarize the latest research progress in the targeted therapy and mechanism of action of SUMO modification pathways in common malignant tumors.
