宏基因组学技术在中医证候研究中的应用与思考

摘　　要：宏基因组学是一种以微生物群落为研究对象的测序研究技术,通过提取特定环境中所有微生物的基因信息,集合分析微生物群落结构、系统进化、代谢路径、基因功能等方面的特征。目前已广泛地用于多种医学相关的微生物研究领域。将宏基因组学引入中医学的研究中,使中医学与同样强调整体性的现代微观技术有机结合,可以弥补中医学研究过程中缺少微观化指标的不足,具有独特的应用前景。中医证候是中医基础理论与中医临床的桥梁,从宏基因组学的研究方法着手,尝试对其进行现代化的定性定量分析,可以部分揭示中医证候的实质,使中医证候更加规范化、科学化。宏基因组学有整体性、动态性、环境统一性的特点,十分符合中医学中“证候”的一般特征。因此,基于宏基因组学技术的中医证候实质研究不仅有较高的可行性,同时可以为中医辨证论治和方剂配伍等理论研究带来新的方向,为推动中医现代化带来新的契机。Metagenomics is a sequencing research technology that takes microbial communities as the research object.It collects and analyzes the characteristics of microbial community structure,system evolution,metabolic pathways,gene functions and other aspects by extracting genetic information of all microorganisms in a specific environment.It has been widely used in a variety of medical-related microbial research fields.Introducing metagenomics into the research of traditional Chinese medicine and effectively combining Chinese medicine with modern micro-technology that also emphasizes integrity can make up for the lack of microscopic indicators in the research process of Chinese medicine,and has a unique application prospect.TCM syndromes are the bridge between the basic theories of TCM and TCM clinics.Starting from the research methods of metagenomics and attempting to carry out modern qualitative and quantitative analysis,it can partially reveal the essence of TCM syndromes and make TCM syndromes more standardized and scientific.Metagenomics has the characteristics of integrity,dynamics,and environmental unity,which is very consistent with the general characteristics of"syndrome"in Chinese medicine.Therefore,research on the essence of TCM syndromes based on metagenomics technology not only has high feasibility,but also can bring new directions for theoretical research such as TCM syndrome differentiation and treatment and prescription compatibility,and bring new opportunities for the modernization of TCM.
