摘 要:为探究食物主要营养成分对花鼠(Tamias sibiricus)肠道菌群结构的影响,以人工饲料饲喂花鼠,以野外自由取食为对照,通过粪便采样,以16S rRNA高通量测序分析花鼠肠道菌群组成。依据食物来源和饲料中脂肪、淀粉含量的不同,将被饲喂的花鼠分为野外组、脂肪组、淀粉组和饲料组。结果显示:不同饲喂条件下,花鼠肠道菌群的优势菌门均为厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)和变形菌门(Proteobacteria),优势菌属为Muribaculaceae_norank和联合乳杆菌属(Ligilactobacillus)。多样性分析显示,野外组肠道菌群物种丰度和多样性较高,但野外组与淀粉组菌群组成相似度较高。相关性分析表明,厚壁菌门与粗蛋白含量呈显著正相关,拟杆菌门与淀粉和粗纤维含量呈显著正相关、与粗脂肪含量呈显著负相关,放线菌门(Actinobacteria)与粗蛋白含量呈显著正相关。研究表明食物成分变化影响了花鼠肠道菌群的组成、结构和丰富度;食物主要营养成分与特定菌群存在显著相关性。In order to investigate the effects of main food nutrients on intestinal flora structure of chipmunks(Tamias sibiri⁃cus),this study used field food as a control and artificially prepared feed to feed chipmunks.Through fecal sampling,16S rRNA high-throughput sequencing was used to determine and study the changes of intestinal flora composition of chip⁃munks.According to the different food sources and the content of fat and starch in the feed,the fed chipmunks were di⁃vided into field group,fat group,starch group and feed group.The results showed that the most abundant bacterial phyla found in chipmunk guts were Firmicutes,Bacteroidetes,and Proteobacteria.The two dominant genera were Muribacula⁃ceae_norank and Ligilactobacillus.The diversity analysis showed that the species abundance and diversity of intestinal flora in the field group were highest.The intestinal flora structure of the two groups was comparable in between the field and the starch group.Correlation analysis showed that crude protein content was positively linked with the presence of Firmicutes.Starch and crude fibre content were positively correlated with Bacteroidetes,whereas crude fat level was inversely corre⁃lated with Bacteroidetes.The presence of Actinobacteria was positively related to the amount of crude protein contents.The above results suggested that the changes in food composition affected the composition,structure and richness of intes⁃tinal flora of chipmunks and that there was significant correlation between food composition and specific intestinal flora.
