摘 要:为分析北京地区猫泛白细胞减少症病毒(Feline panleukopenia virus,FPV)的流行毒株及其VP2基因遗传变异情况,对FPV胶体金试纸条检测结果为阳性的病死猫组织进行病毒分离,通过细胞病变、荧光定量PCR、间接免疫荧光和全基因组测序等试验,分离鉴定到1株FPV,命名为FPVBJ-CP/2022株。对决定病毒宿主范围和抗原性的VP2基因系统进化树分析表明,分离株与FPVJF-3株、FPVBeijing-01/2018聚为一支,与FPV疫苗株、CPV群亲缘关系较远。VP2蛋白主要功能位点分析结果显示,VP2蛋白中决定病毒抗原性和宿主嗜性的10个关键位点氨基酸均未发生突变,但该分离株的VP2蛋白序列与GenBank其他FPV毒株的VP2蛋白相比,91位氨基酸由丙氨酸(A)突变为丝氨酸(S),505位氨基酸由天冬酰胺(N)突变为天冬氨酸(D),该突变是否对FPVBJ-CP/2022株VP2蛋白的功能和生物学特性造成改变,有待于进一步研究。论文通过对毒株的遗传变异分析,为更好地预防和控制FPV感染提供了研究基础。To analyze the prevalent strain and the genetic variation of VP2 gene of feline panleukopenia virus(FPV)in Beijing area,in this study,pathological tissues were collected from a dead cat with positive results of FPV by colloidal gold test in Beijing and were used to isolate FPV by F81 cells.One FPV strain was isolated and identified by cytopathic test,fluorescence quantitative PCR,indirect immunofluorescence assay and whole genome sequencing.The isolated virus strain was named FPV BJ-CP/2022 strain.Phylogenetic tree of VP2 gene which determines host range and antigenicity of the virus,showed that FPV BJ-CP/2022 strain,FPV JF-3strain,FPV Beijing-01/2018 strain were on the same small branch,while had a distant relationship with FPV vaccine strain and CPV group.The analysis of VP2 protein main function sites showed that there were no mutations in the lo key amino acid sites of VP2 protein,which determine the antigenicity and host property of parvovirus.However,compared with other FPV strains in GenBank,amino acid position in the VP2 protein of this virus stain was mutated from alanine(A)to serine(S)at amino acid position 91,and from asparagine(N)to aspartic acid(D)at amino acid position 505.Whether this mutation changes the function and biological characteristics of FPV BJ-CP/2022 strain VP2 protein remains to be further studied.In this study,the genetic variation analysis of the isolated virus strain provided research basis for better prevention and control of FPV infection in the next work.
