摘  要:目的探讨白细胞介素9(IL-9)对小鼠肺腺癌生长的影响及其可能的作用机制。方法将小鼠肺腺癌CMT167细胞接种于BALB/C小鼠皮下,构建小鼠肺腺癌皮下移植瘤模型,观察IL-9对小鼠肿瘤生长的影响。采用流式细胞术(FCM)和蛋白免疫印迹法(Western blot)检测小鼠肺腺癌CMT167细胞IL-9受体(IL-9R)的表达情况。不同浓度IL-9体外作用于CMT167细胞后,观察不同时间点其细胞活力和细胞凋亡的变化。采用FCM法分析小鼠肿瘤组织中骨髓来源免疫抑制性细胞(myeloid-derived immunosuppressive cells,MDSCs)、CD4^(+)T细胞和CD8^(+)T细胞百分比的变化情况。结果与模型对照组小鼠相比,IL-9能够明显抑制小鼠肺腺癌皮下移植瘤的生长,并减轻移植瘤的重量(均P<0.01)。小鼠肺腺癌CMT167细胞中IL-9R呈阳性表达。不同浓度IL-9体外作用小鼠肺腺癌CMT167细胞后其细胞活力和凋亡均无明显变化(均P>0.05)。IL-9降低了小鼠肿瘤组织中MDSCs的数量(P<0.05),同时增加了肿瘤组织中CD4^(+)T细胞和CD8^(+)T细胞的比例(P<0.01)。结论IL-9能够抑制小鼠肺腺癌移植瘤的生长,这可能与IL-9降低小鼠肿瘤组织中MDSCs的数量,增加CD4^(+)T细胞和CD8^(+)T细胞比例有关。Objective To investigate the effect of interleukin-9(IL-9)on the growth of lung adenocarcinoma in mice and its possible mechanism of action.Methods Mouse lung adenocarcinoma CMT167 cells were inoculated subcutaneously in BALB/C mice to construct a subcutaneous transplantation tumor model of mouse lung adenocarcinoma and to observe the effect of IL-9 on the growth of mouse tumors.Flow cytometry(FCM)and Western blot(WB)were used to detect the expression of IL-9 receptor(IL-9R)in mouse lung adenocarcinoma CMT167 cells.CMT167 cells were treated with different concentrations of IL-9 in vitro,and changes in cell viability and apoptosis were observed at different time points.Changes in the percentages of myeloid-derived immunosuppressive cells(MDSCs),CD4^(+)T cells,and CD8^(+)T cells in the tumor tissues of mice were analyzed by using FCM.Results Compared with mice in control group,the growth of mouse lung subcutaneous graft tumors was significantly inhibited,and the weight of graft tumor was reduced after IL-9 treatment(both P<0.01).Compared with the control group,the expression of IL-9R in mouse lung adenocarcinoma CMT167 cells was significantly increased(P<0.01).After treatment with different concentrations of IL-9 in vitro,there were no significant changes in the viability and apoptosis of mouse lung adenocarcinoma CMT167 cells compared to the control group.The number of MDSCs in mouse tumor tissues was decreased after IL-9 treatment(P<0.05),while proportions of CD4^(+)T cells and CD8^(+)T cells in tumor tissues were increased(P<0.01).Conclusion IL-9Reduces the number of MDSCs and increases the proportions of CD4^(+)T cells and CD8^(+)T cells in mouse tumor tissues,therefore inhibits the growth of lung adenocarcinoma tumors in mice.
