摘 要:目的探讨长链非编码RNA(lncRNA)在浅表性食管鳞状细胞癌中表达及其临床意义。方法选取2022年6月到2023年6月新乡市中心医院和上海市胸科医院收治的54例浅表性食管鳞状细胞癌和癌旁组织作为研究对象,采用lncRNA芯片分析癌旁组织和浅表性食管鳞状细胞癌差异表达lncRNA,分析差异表达基因的功能富集和生物学调控通路。选择差异最为显著的lncRNANORAD作为研究对象,采用荧光定量分析癌旁组织和浅表性食管鳞状细胞癌组织中lncRNANORAD表达水平。分析lncRNANORAD表达水平与浅表性食管鳞状细胞癌患者临床病理特征和预后的关系。采用多因素回归分析分析浅表性食管鳞状细胞癌预后的独立因素。计量数据比较采用t检验。结果采用lncRNA芯片共筛选出2180条差异表达的lncRNA(上调lncRNA 1432条,下调lncRNA748条)。上调倍数最大的是lncRNANORAD(12倍)。京都基因与基因组百科全书(KEGG)富集显示这些lncRNA参与诸多生物代谢和信号转导相关的通路,包括细胞糖代谢过程、细胞生长和死亡、蛋白质相互作用、蛋白质转录翻译后修饰、DNA损伤修复和氨基酸嘌呤代谢。癌旁组织中lncRNANORAD表达水平(1.01±0.17)明显低于浅表性食管鳞状细胞癌(1.95±0.17),差异有统计学意义(t=29.150,P<0.05)。中高分化患者浅表性食管鳞状细胞癌组织lncRNANORAD表达水平(1.85±0.12)明显低于低分化患者(2.07±0.15),差异有统计学意义(t=5.828,P<0.05)。管腔狭窄患者浅表性食管鳞状细胞癌组织lncRNANORAD表达水平(2.03±0.14)明显高于无管腔狭窄患者(1.88±0.16)。差异有统计学意义(t=3.592,P<0.05)。有脉管癌栓患者浅表性食管鳞状细胞癌组织lncRNANORAD表达水平(2.05±0.17)明显高于无脉管癌栓患者(1.90±0.13),差异有统计学意义(t=5.248,P<0.05)。淋巴结转移患者浅表性食管鳞状细胞癌组织lncRNANORAD表达水平(1.83±0.13)明显高于未见淋巴结转移患者(2.0Objective To investigate the expression of long non-coding RNA(lncRNA)in superfi-cial esophageal squamous cell carcinoma and its clinical significance.Methods A total of 54 cases of superficial esophageal cancer and adjacent tissues treated from June 2022 to June 2023 were selected as re-search objects in Xinxiang Central Hospital and Shanghai Chest Hospital.LncRNA microarray was used to analyze lncRNA differentially expressed in adjacent tissues and tumor tissues,and the functional enrich-ment and biological regulatory pathways of differentially expressed genes were analyzed.LncRNA NORAD with the most significant difference was selected as the study objects,and the expression level of lncRNA NORAD in adjacent and superficial esophageal cancer tissues was quantitatively analyzed by fluorescence.The relationship between lncRNA NORAD expression level and clinicopathological features and prognosis of patients with superficial esophageal cancer was analyzed.Multivariate regression analysis was used to analyze the independent prognostic factors of superficial esophageal cancer.Measurement data were compared by t test.Results In this study,a total of 2180 differentially expressed lncRNAs were screened using lncRNA chips(1432 lncRNAs up-regulated and 748 lncRNAs down-regulated).LncRNA NORAD had the largest up-regulation ratio(12 times).Kyoto encyclopedia of genes and genomes(KEGG)enrichment showed that these lncRNAs were involved in many biological metabolic and signal transduction related pathways,including cellular glucose metabolism,cell growth and death,protein interactions,protein transcriptional post-translational modification,DNA damage repair,and amino acid purine metabolism.The expression level of lncRNA NORAD in adjacent tissues(1.O1±0.17)was significantly lower than that in superficial tumor cells(1.95±0.17,t=29.150,P<0.05).LncRNA NORAD expression level in superficial tumor tissues of medium-high differentiation patients(1.85±0.12)was significantly lower than that of lowdifferentiation patients(2.07±0.15,
